Abstract

Dalam memproduksi biodiesel dari Crude Plam Oil menimbulkan tantangan berupa kandungan asam lemak bebas (FFA) yang tinggi sehingga kurang sesuai untuk proses transesterifikasi katalis basa konvensional, sedangkan katalis kimia asam maupun basa menimbulkan masalah pemurnian, pembentukan sabun, dan konsumsi energi yang relatif tinggi. Imobilisasi sel utuh R oryzae dipilih sebagai alternatif lipase murni untuk menurunkan biaya proses sekaligus meningkatkan stabilitas biokatalis dan potensi pemakaian berulang pada sistem produksi biodiesel. Penelitian ini bertujuan untuk mengevaluasi penggunaan biokatalis enzimatik dari sel utuh R oryzae yang diimobilisasi pada matriks PVA-Alginat termodifikasi selulosa nanokristal CNC-Biochar untuk produksi biodiesel dari Crude Palm Oil. Metode yang digunakan meliputi pembuatan bead gel PVA–Alginat/CNC–biochar berisi sel R. oryzae, karakterisasi menggunakan FTIR, SEM, untuk mengamati gugus fungsi, morfologi, serta uji esterifikasi–transesterifikasi CPO dengan metanol dalam sistem batch diikuti analisis kadar biodiesel menggunakan GC. Hasil yang diharapkan adalah terbentuknya bead gel dengan kekuatan mekanik yang baik, distribusi sel merata, permukaan lebih hidrofobik, serta diperolehnya yield biodiesel tinggi dan stabilitas biokatalis yang memungkinkan penggunaan ulang sehingga berpotensi menjadi teknologi produksi biodiesel yang lebih efisien dan berkelanjutan. Berdasarkan penelitian yang dilakukan di dapatkan hasil bahwa karakterisasi FTIR menunjukkan bahwa bead gel PVA–Alginat–CNC–Biochar berhasil terbentuk melalui mekanisme double crosslinking yang melibatkan ikatan kovalen, ionik, dan hidrogen, sedangkan hasil SEM mengonfirmasi keberhasilan imobilisasi Rhizopus oryzae. Rasio matriks CNC 3:2 menghasilkan keseimbangan sifat hidrofilik–hidrofobik yang optimal sehingga memberikan performa biokatalis terbaik. Dosis metanol optimum yang dapat ditoleransi biokatalis adalah rasio 1:12, sedangkan peningkatan rasio hingga 1:18 menurunkan yield dan pada rasio 1:21 tidak terbentuk biodiesel. Yield biodiesel tertinggi yang diperoleh mencapai 70% pada rasio metanol 1:12 dengan matriks CNC 3:2. Biokatalis terimobilisasi juga dapat digunakan kembali hingga tiga siklus dengan hasil biodiesel berturut-turut sebesar 38 mL, 23 mL, dan 13 mL, sedangkan pada siklus keempat biodiesel tidak terbentuk.

Biodiesel production from Crude Palm Oil (CPO) faces challenges due to its high free fatty acid (FFA) content, making it less suitable for conventional base-catalyzed transesterification. In addition, acid- and base-catalyzed chemical processes present drawbacks, including complex purification, soap formation, and high energy consumption. Whole-cell immobilization of Rhizopus oryzae was selected as an alternative to purified lipase to reduce production costs while improving biocatalyst stability and reusability. This study aimed to evaluate the application of immobilized whole-cell R. oryzae on a PVA–alginate matrix modified with cellulose nanocrystals (CNC) and biochar for biodiesel production from Crude Palm Oil. The methodology included the preparation of PVA–alginate/CNC–biochar gel beads containing R. oryzae cells, characterization using FTIR and SEM to analyze functional groups and morphology, followed by batch esterification–transesterification of CPO with methanol and biodiesel quantification using gas chromatography (GC). FTIR characterization confirmed the successful formation of PVA–alginate–CNC–biochar gel beads through a double-crosslinking mechanism involving covalent, ionic, and hydrogen bonding, while SEM analysis verified the successful immobilization of R. oryzae. A CNC:biochar ratio of 3:2 provided the optimum hydrophilic–hydrophobic balance, resulting in the best biocatalyst performance. The optimum methanol-to-oil molar ratio tolerated by the biocatalyst was 1:12, whereas increasing the ratio to 1:18 reduced biodiesel yield and no biodiesel was produced at a ratio of 1:21. The highest biodiesel yield reached 70% at a methanol ratio of 1:12 using a CNC:biochar matrix ratio of 3:2. Furthermore, the immobilized biocatalyst remained reusable for up to three cycles, producing 38 mL, 23 mL, and 13 mL of biodiesel in the first, second, and third reuse cycles, respectively, while no biodiesel was obtained in the fourth cycle
