Abstract

Luka yang terinfeksi Staphylococcus aureus USA300 dapat mengalami keterlambatan penyembuhan akibat inflamasi berkepanjangan yang mengganggu regenerasi jaringan. SKLY1-SC10 merupakan formulasi poliherbal yang mengandung ekstrak bangle (Zingiber montanum), lidah buaya (Aloe vera), dan jambu mete (Anacardium occidentale) yang berpotensi mempercepat penyembuhan luka terinfeksi. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian SKLY1-SC10 terhadap penyembuhan luka yang diinfeksi S. aureus USA300 pada mencit (Mus musculus). Mencit dilukai menggunakan skin biopsy punch, diinfeksi S. aureus USA300, kemudian diberi perlakuan berupa SKLY1-SC10 5%, SKLY1-SC10 10%, obat komersil, solvent, dan kontrol negatif. Kelompok tidak dilukai (sehat) digunakan sebagai kontrol normal. Parameter yang diamati meliputi luas luka, ketebalan epidermis, skor histopatologi, dan jumlah fibroblas. Data dianalisis menggunakan One Way ANOVA dan uji Tukey. Hasil penelitian menunjukkan bahwa luas luka pada hari ke-4 dan ke-6 berbeda signifikan antar kelompok. Kelompok SKLY1-SC10 5% dan 10% berada pada kelompok yang sama dengan obat komersil dan memiliki luas luka yang lebih kecil dibandingkan kelompok solvent dan kontrol negatif. Ketebalan epidermis kelompok SKLY1-SC10 tidak berbeda signifikan dengan kelompok sehat, sedangkan kelompok solvent dan kontrol negatif berbeda signifikan dengan kedua kelompok tersebut. Hasil skoring histopatologi menunjukkan bahwa kelompok SKLY1-SC10 10% memperoleh skor tertinggi sebesar 11,5 dan mendekati kelompok tidak dilukai (sehat). Analisis jumlah fibroblas juga menunjukkan bahwa kelompok SKLY1-SC10 10% berada pada kelompok yang sama dengan kelompok tidak dilukai (sehat). Kondisi tersebut menunjukkan bahwa jaringan telah mengalami maturasi yang lebih baik, ditandai dengan penurunan jumlah fibroblas yang disertai peningkatan deposisi dan maturasi serabut kolagen. Berdasarkan hasil penelitian dapat disimpulkan bahwa pemberian SKLY1-SC10, khususnya pada konsentrasi 10%, mampu mempercepat penutupan luka yang diinfeksi S. aureus USA300 dan mendorong proses penyembuhan menuju fase remodeling awal.

Wounds infected with Staphylococcus aureus USA300 may experience delayed healing due to prolonged inflammation that disrupts tissue regeneration. SKLY1-SC10 is a polyherbal formulation containing extracts of bangle (Zingiber montanum), aloe vera (Aloe vera), and cashew (Anacardium occidentale), which has the potential to accelerate the healing of infected wounds. This study aimed to evaluate the effect of SKLY1-SC10 on the healing of S. aureus USA300-infected wounds in mice (Mus musculus). Wounds were created using a skin biopsy punch and subsequently infected with S. aureus USA300. The mice were then treated with SKLY1-SC10 at concentrations of 5% and 10%, a commercial drug, solvent, or left untreated as a negative control. An uninjured (healthy) group was used as the normal control. The observed parameters included wound area, epidermal thickness, histopathological score, and fibroblast count. Data were analyzed using One-Way ANOVA followed by Tukey’s post hoc test. The results showed significant differences in wound area among groups on days 4 and 6. The SKLY1-SC10 5% and 10% groups were classified in the same Tukey grouping as the commercial drug group and exhibited smaller wound areas than the solvent and negative control groups. Epidermal thickness in the SKLY1-SC10 groups was not significantly different from that of the healthy group, whereas the solvent and negative control groups differed significantly from both the healthy and SKLY1-SC10 groups. Histopathological scoring revealed that the SKLY1-SC10 10% group achieved the highest score (11.5), closely resembling the healthy group. Fibroblast analysis also showed that the SKLY1-SC10 10% group belonged to the same Tukey grouping as the healthy group. These findings indicate improved tissue maturation, characterized by a reduced fibroblast count accompanied by increased collagen deposition and maturation. It can be concluded that SKLY1-SC10, particularly at a concentration of 10%, effectively accelerates the closure of S. aureus USA300-infected wounds and promotes the wound healing process toward the early remodeling phase.
