Abstract
Meningkatnya resistensi antimikroba dan kemampuan pembentukan biofilm oleh Candida albicans menjadi tantangan utama dalam pengendalian infeksi karena biofilm mampu melindungi sel mikroba dari berbagai agen antimikroba konvensional. Salah satu pendekatan yang menjanjikan adalah pengembangan nanomaterial berbasis sintesis hijau yang memiliki aktivitas antimikroba dan berpotensi diaplikasikan dalam photodynamic inactivation (PDI). Penelitian ini bertujuan mensintesis nanopartikel Ag@AgCl menggunakan ekstrak daun nilam (Pogostemon cablin Benth.) sebagai agen pereduksi dan penstabil alami, dikonjugasikan dengan kurkumin, serta mengevaluasi karakteristik fisikokimia, aktivitas antimikroba, dan efektivitas PDI terhadap biofilm Candida albicans. Karakterisasi nanopartikel dilakukan untuk mengkarakterisasi karakteristik sifat optik, gugus fungsi, struktur kristal, morfologi, komposisi unsur, dan distribusi ukuran partikel menggunakan spektroskopi UV–Vis, FTIR, XRD, FESEM, EDX, dan PSA. Aktivitas antimikroba diuji dengan metode difusi cakram, sedangkan efektivitas PDI terhadap biofilm dianalisis menggunakan XTT assay.Hasil skrining fitokimia menunjukkan adanya flavonoid, alkaloid, terpenoid, dan tanin yang berperan dalam proses sintesis nanopartikel. Karakterisasi UV–Vis menunjukkan puncak serapan maksimum (λmax) pada panjang gelombang 442 nm dengan energi celah pita 2,29 eV, sedangkan analisis XRD mengonfirmasi terbentuknya fase kristalin Ag dan AgCl dengan ukuran kristalit 6,9-18,5 nm. Ukuran partikel Ag@AgCl-NPs meningkat dari 92,01 nm menjadi 199,17 nm setelah konjugasi dengan kurkumin, yang didukung oleh perubahan gugus fungsi pada spektrum FTIR. Di antara seluruh sampel yang diuji, Ag@AgCl-NPs menghasilkan diameter zona hambat terbesar terhadap Staphylococcus aureus dan Candida albicans, masing-masing sebesar 5,27 mm dan 8,58 mm. Efektivitas inaktivasi biofilm C. albicans mencapai (87,59 ±12,44)% dan meningkat menjadi (92,21 ± 2,41)% setelah perlakuan photodynamic inactivation (PDI). Meskipun konjugasi dengan kurkumin berhasil memodifikasi sifat fisikokimia nanopartikel, aktivitas antimikrobanya menurun dibandingkan dengan Ag@AgCl-NPs tanpa konjugasi. Oleh karena itu, Ag@AgCl-NPs hasil sintesis hijau tanpa konjugasi kurkumin berpotensi sebagai agen antimikroba dan fotodinamik yang efektif untuk pengendalian biofilm Candida albicans.

The increasing prevalence of antimicrobial resistance and the ability of Candida albicans to form biofilms have become major challenges in infection control, as biofilms protect microbial cells from conventional antimicrobial agents. One promising strategy to address this issue is the development of green-synthesized nanomaterials with antimicrobial properties and potential applications in photodynamic inactivation (PDI). This study aimed to synthesize Ag@AgCl nanoparticles using Pogostemon cablin Benth. leaf extract as a natural reducing and stabilizing agent, conjugate them with curcumin, and evaluate their physicochemical characteristics, antimicrobial activity, and PDI efficacy against Candida albicans biofilms. The nanoparticles were characterized to determine their optical properties, functional groups, crystal structure, morphology, elemental composition, and particle size distribution using UV–Vis spectroscopy, FTIR, XRD, FESEM, EDX, and PSA. Antimicrobial activity was evaluated using the disk diffusion method, while the antibiofilm efficacy under PDI was assessed using the XTT assay. Phytochemical screening revealed the presence of flavonoids, alkaloids, terpenoids, and tannins, which contributed to nanoparticle biosynthesis. UV–Vis analysis showed a surface plasmon resonance (SPR) peak at 442 nm with a band gap energy of 2.29 eV, while XRD confirmed the formation of crystalline Ag and AgCl phases with crystallite sizes ranging from 6.9 to 18.5 nm. The average particle size increased from 92,01 nm for Ag@AgCl nanoparticles to 199,17 nm after curcumin conjugation, which was further supported by changes in the FTIR spectra. Ag@AgCl nanoparticles exhibited the highest antimicrobial activity against Staphylococcus aureus and Candida albicans, with inhibition zone diameters of 5.27 mm dan 8.58, respectively. The nanoparticles achieved a biofilm inactivation efficiency of (87.59 ± 12.44)%, which increased to (92.21 ± 2.41)% following PDI treatment. Although curcumin conjugation successfully modified the physicochemical properties of the nanoparticles, it resulted in reduced antimicrobial activity compared with unconjugated Ag@AgCl nanoparticles. Therefore, green-synthesized Ag@AgCl nanoparticles without curcumin conjugation demonstrated superior antimicrobial and antibiofilm performance and are considered more effective for controlling Candida albicans biofilms.
