Abstract
Industri pengolahan agar berbahan baku Gracilaria sp. menghasilkan limbah cair dengan kandungan organik tinggi yang berpotensi mencemari ekosistem pesisir apabila tidak dikelola dengan baik. Di sisi lain, kebutuhan terhadap docosahexaenoic acid (DHA) terus meningkat sehingga diperlukan sumber produksi alternatif yang lebih berkelanjutan. Penelitian ini bertujuan mengkaji potensi limbah cair industri agar Gracilaria sp. sebagai sumber karbon alternatif untuk produksi DHA melalui kultivasi heterotrof mikroorganisme laut, yaitu Thraustochytrids dan Crypthecodinium cohnii. Metode penelitian meliputi isolasi dan identifikasi mikroorganisme, karakterisasi limbah cair berdasarkan parameter TOC, pH, dan salinitas, kultivasi heterotrof pada variasi konsentrasi TOC, analisis pertumbuhan biomassa, ekstraksi lipid, serta analisis profil asam lemak menggunakan GC. Hasil penelitian menunjukkan bahwa limbah cair industri agar Gracilaria sp. mampu dimanfaatkan sebagai media kultivasi heterotrof untuk mendukung pertumbuhan biomassa dan produksi DHA. Perbedaan karakteristik media memengaruhi kemampuan produksi DHA pada kedua mikroorganisme. Thraustochytrids menunjukkan kinerja lebih tinggi dibandingkan Crypthecodinium cohnii pada seluruh perlakuan. Produksi DHA tertinggi diperoleh pada TOC 2300 Thraustochytrids dan 11,792 µg/g untuk C. cohnii. Hasil ini menunjukkan bahwa Thraustochytrids memiliki kemampuan adaptasi dan pemanfaatan nutrien yang lebih baik sehingga lebih potensial sebagai agen biokonversi limbah cair industri agar menjadi produk DHA bernilai tambah.

The agar processing industry using Gracilaria sp. as raw material generates liquid waste with high organic content, which may cause coastal environmental problems if not properly managed. Meanwhile, the increasing demand for docosahexaenoic acid (DHA) requires the development of sustainable alternative production sources. This study aimed to evaluate the potential of Gracilaria sp. agar industry liquid waste as an alternative carbon source for DHA production through heterotrophic cultivation of marine microorganisms, namely Thraustochytrids and Crypthecodinium cohnii. The methods included microorganism isolation and identification, liquid waste characterization based on TOC, pH, and salinity, heterotrophic cultivation at different TOC concentrations, biomass growth analysis, lipid extraction, and fatty acid profile analysis using GC. The results showed that Gracilaria sp. agar industry liquid waste could be utilized as a heterotrophic cultivation medium to support biomass growth and DHA production. Differences in medium characteristics affected DHA productivity of both microorganisms. Thraustochytrids showed higher DHA production performance than Crypthecodinium cohnii in all treatments. DHA production of Thraustochytrids at TOC 2300, for Thraustochytrids and 11.792 µg/g for C. cohnii. These results indicate that Thraustochytrids have better adaptation and nutrient utilization capability, making them more promising as bioconversion agents for converting agar industry liquid waste into value-added DHA products.
