Abstract
Susu adalah produk pangan bernutrisi tinggi yang rentan terhadap adulterasi berupa penambahan air. Deteksi pengenceran secara konvensional umumnya menggunakan metode destruktif atau memerlukan reagen kimia, sehingga diperlukan alternatif yang lebih praktis dan non-destruktif. Penelitian ini mengkaji pengaruh konsentrasi susu UHT full cream terhadap pola spekel laser melalui pendekatan pengolahan citra digital. Sampel diencerkan dengan air deionisasi pada variasi konsentrasi susu 10% hingga 100% dengan interval 10%. Laser He-Ne (λ = 632,8 nm) diarahkan ke sampel dalam kuvet kuarsa, dan pola spekel direkam pada sudut 90° menggunakan sensor CMOS webcam beresolusi 1920×1080 piksel. Data direkam dalam format video selama 1 menit, kemudian diolah menggunakan MATLAB dengan mengekstrak ±680 frame per sampel pada rentang detik ke-11 hingga ke-50. Setiap frame menjalani normalisasi Min-Max sebelum dihitung dua parameter statistik spekel secara spasial, yaitu kontras spekel dan mean intensity. Hasil penelitian menunjukkan bahwa kontras spekel meningkat secara linear seiring bertambahnya konsentrasi susu, dari 0,062573 pada konsentrasi 10% hingga 0,075657 pada konsentrasi 100% (R² = 0,968), sedangkan mean intensity menurun secara linear dari 0,714817 menjadi 0,671657 (R² = 0,914). Peningkatan kontras spekel dijelaskan melalui mekanisme hamburan Mie oleh globula lemak dan misel kasein, sedangkan penurunan mean intensity dikaitkan dengan atenuasi foton akibat hamburan multipel yang sesuai dengan hukum Beer-Lambert. Penelitian ini membuktikan bahwa metode spekel laser berbasis pengolahan citra digital mampu mengkarakterisasi konsentrasi susu UHT full cream secara kuantitatif dan non-destruktif dengan konfigurasi instrumen sederhana tanpa algoritma machine learning.

Milk is a highly nutritious food product particularly vulnerable to adulteration through water addition. Conventional detection methods are generally destructive or require chemical reagents, necessitating a more practical and non-destructive alternative. This study investigates the effect of UHT full cream milk concentration on laser speckle patterns through a digital image processing approach. Samples were diluted with deionized water at milk concentrations ranging from 10% to 100% with 10% intervals. A He-Ne laser (λ = 632.8 nm) was directed at samples in a quartz cuvette, and speckle patterns were recorded at 90° using a lensless CMOS webcam sensor at 1920×1080 pixel resolution. Data were recorded as video files and processed in MATLAB by extracting approximately 680 frames per sample from the 11th to 50th second. Each frame underwent Min-Max normalization prior to spatial computation of two statistical speckle parameters: speckle contrast and mean intensity. Results show that speckle contrast increased linearly from 0.062573 at 10% to 0.075657 at 100% (R² = 0.968), while mean intensity decreased linearly from 0.714817 to 0.671657 (R² = 0.914). The increase in speckle contrast is attributed to Mie scattering by fat globules and casein micelles, while the decrease in mean intensity is consistent with photon attenuation from multiple scattering following the Beer-Lambert law. This study demonstrates that a laser speckle method based on digital image processing can quantitatively and non-destructively characterize UHT full cream milk concentration using a simple instrumental configuration without machine learning algorithms.
