Abstract
Plant growth promoting rhizobacteria (PGPR) merupakan bakteri yang telah banyak dilaporkan sebagai agen biokontrol. Saat ini, Laboratorium Mikrobiologi dan Bioteknologi memiliki koleksi isolat yang diisolasi dari tanah terinfeksi G. boninense. Untuk mengetahui potensi biokontrol tersebut, penelitian ini dilakukan untuk menghambat yaitu Xanthomonas oryzae pv. oryzae (Xoo) dan Ganoderma boninense. Penelitian ini memiliki tujuan, pertama untuk mengeksplorasi kemampuan isolat koleksi dalam menghasilkan biosurfaktan. Kedua, menguji aktivitas antimikroba terhadap Xoo dan G. boninense. Ketiga, mendeteksi keberadaan gen pengkode biosurfaktan pada isolat potensial. Penelitian diawali dari skrining aktivitas antagonistik dan deteksi kualitatif biosurfaktan. Selanjutnya, isolat bakteri koleksi yang berpotensi menghasilkan biosurfaktan, diuji silang terhadap bakteri patogen Xoo dan G. boninense, serta deteksi gen biosurfaktan menggunakan 4 primer spesifik. Terakhir, isolat potensial dikarakterisasi dengan gen 16S rRNA. Data dianalisis secara kuantitatif menggunakan uji ANOVA one way. Hasil penelitian menunjukkan bahwa dari 16 isolat bakteri koleksi, sebanyak sembilan isolat mampu menghasilkan biosurfaktan kasar. Produksi biosurfaktan kasar tertinggi diperoleh pada isolat 0S2 sebesar 2,64 g/L dan isolat 2N4 sebesar 2,41 g/L. Biosurfaktan kasar isolat 2S5 dan 2N6 menunjukkan aktivitas antibakteri yang signifikan terhadap Xoo dengan diameter zona inhibisi masing-masing sebesar 15,216 ± 1,272 mm dan 12,088 ± 1,516 mm. Pada pengujian terhadap G. boninense, supernatan biosurfaktan seluruh isolat mampu menghambat pertumbuhan miselium selama tujuh hari inkubasi, dengan aktivitas penghambatan tertinggi ditunjukkan oleh isolat 2N4 dan 2S2 yang memiliki nilai Percentage Inhibition of Radial Growth (PIRG) masing-masing sebesar 32,254% dan 31,570%. Deteksi gen menggunakan empat primer biosurfaktan menunjukkan adanya gen yang berhasil teramplifikasi pada masing-masing isolat. Hasil crossing primer menunjukkan bahwa tidak semua isolat memiliki gen pengkode jenis biosurfaktan yang terdeteksi. Isolat yang menghasilkan amplikon pada crossing primer mengindikasikan potensi keberadaan gen pengkode biosurfaktan yang masih memerlukan konfirmasi lebih lanjut melalui analisis molekuler.

Plant growth promoting rhizobacteria (PGPR) is a bacterium that has been widely reported as a biocontrol agent. Currently, the Laboratory of Microbiology and Biotechnology has a collection of isolates isolated from G. boninense-infected soils. To find out the potential for biocontrol, this study was carried out to inhibit Xanthomonas oryzae pv. oryzae (Xoo) and Ganoderma boninense. This research has the objective, first, to explore the ability of collection isolates in producing biosurfactants. Second, to test the antimicrobial activity of Xoo and G. boninense. Third, detect the presence of biosurfactant-coding genes in potential isolates. The research began with the screening of antagonistic activity and qualitative detection of biosurfactants. Furthermore, the isolates of collected bacteria that have the potential to produce biosurfactants were cross-tested against pathogenic bacteria Xoo and G. boninense, as well as the detection of biosurfactant genes using 4 specific primers. Finally, the potential isolates were characterized by the 16S rRNA gene. The data was analyzed quantitatively using the one-way ANOVA test. The results showed that out of 16 bacterial isolates collected, as many as nine isolates were able to produce coarse biosurfactants. The highest coarse biosurfactant production was obtained in 0S2 isolate of 2.64 g/L and 2N4 isolate of 2.41 g/L. Coarse biosurfactants of 2S5 and 2N6 isolates showed significant antibacterial activity against Xoo with inhibition zone diameters of 15.216 ± 1.272 mm and 12.088 ± 1.516 mm, respectively. In testing on G. boninense, the biosurfactant supernatant of the whole isolate was able to inhibit the growth of the mycelium during seven days of incubation, with the highest inhibition activity shown by 2N4 and 2S2 isolates which had Percentage Inhibition of Radial Growth (PIRG) values of 32.254% and 31.570%, respectively. Gene detection using four biosurfactant primers showed the presence of genes that were successfully amplified in each isolate. The results of primary crossing showed that not all isolates had a biosurfactant-type encoding gene detected. The isolate that produces amplicons at primary crossing indicates the potential presence of biosurfactant-coding genes that still require further confirmation through molecular analysis.
