Abstract
Porang (Amorphophallus muelleri Blume) merupakan tanaman bernilai ekonomi tinggi karena kandungan glukomanan dapat dimanfaatkan dalam industri pangan dan kesehatan. Namun, ketersediaan lahan optimal semakin berkurang akibat alih fungsi dan aktivitas tambang yang meninggalkan residu logam berat seperti timbal (Pb). Timbal diketahui dapat menghambat pertumbuhan serta menyebabkan terjadinya respon adaptasi dari segi morfologi dan anatomi. Penelitian ini bertujuan mengkaji respon morfologi dan anatomi porang terhadap cekaman Pb(NO₃)₂. Penelitian dilaksanakan di Greenhouse Eco Urban Farming ITS menggunakan rancangan acak lengkap (RAL) menggunakan lima perlakuan konsentrasi Pb(NO₃)₂ (0, 150, 300, 450, dan 600 ppm) dengan ulangan masing-masing konsentrasi sebanyak lima kali. Parameter yang diamati meliputi morfologi tinggi tanaman, luas daun, berat segar umbi, diameter umbi, dan pengukuran panjang akar, serta parameter anatomi yaitu kerapatan serta indeks stomata, dan penampang melintang akar dan umbi. Data dianalisis menggunakan ANOVA One Way (α ≤ 0,05) satu arah dan dilakukan uji lanjut menggunakan uji Tukey dengan selang kepercayaan 95%. Respons morfologi paparan cekaman logam Pb(NO3)2 memberikan pengaruh nyata terhadap parameter tinggi tanaman di konsentrasi 300 sampai 600 ppm (63,4 cm) serta luas daun pada konsentrasi 450 sebesar 950,2 mm2 dan 600 sebesar 850,6 mm2. Sebaliknya, paparan cekaman logam Pb(NO3)2 tidak memberikan pengaruh nyata terhadap panjang akar, diameter umbi, berat segar umbi. Respons anatomi paparan cekaman logam Pb(NO3)2 memberikan pengaruh nyata terhadap parameter persentase stomata menutup meningkat mencapai 74,36% pada konsentrasi 600 ppm, serta kerapatan stomata konsentrasi 150 sampai 300 ppm (72,65 stomata/mm2). Namun, pemberian cekaman tidak berpengaruh pada anatomi akar meliputi ketebalan epidermis, korteks, jaringan pembuluh, serta tidak berpengaruh pada kerapatan idioblas glukomanan di umbi.

Porang (Amorphophallus muelleri Blume) is a plant with high economic value because the glucomannan content can be utilized in the food and health industries. However, the availability of optimal land is decreasing due to land conversion and mining activities that leave heavy metal residues such as lead (Pb). Lead is known to inhibit growth and cause adaptive responses in terms of morphology and anatomy. This study aims to examine the morphological and anatomical responses of porang to Pb(NO₃)₂ stress. The study was conducted at the ITS Eco Urban Farming Greenhouse using a completely randomized design (CRD) using five Pb(NO₃)₂ concentration treatments (0, 150, 300, 450, and 600 ppm) with five replications of each concentration. The parameters observed included morphological plant height, leaf area, tuber fresh weight, tuber diameter, and root length measurements, as well as anatomical parameters, namely stomatal density and index, and root and tuber cross-sections. Data were analyzed using one-way ANOVA (α ≤ 0.05) and further tests were carried out using the Tukey test with a 95% confidence interval. The morphological response of Pb(NO₃)₂ metal stress exposure had a significant effect on plant height parameters at concentrations of 300 to 600 ppm (63.4 cm) and leaf area at concentrations of 450 ppm of 950.2 mm2 and 600 of 850.6 mm2. In contrast, exposure to Pb(NO₃)₂ metal stress did not have a significant effect on root length, tuber diameter, and tuber fresh weight. The anatomical response of Pb(NO₃)₂ metal stress exposure had a significant effect on the parameters of the percentage of stomata closing, increasing to 74.36% at a concentration of 600 ppm, as well as stomatal density at concentrations of 150 to 300 ppm (72.65 stomata/mm2). However, stress administration did not affect root anatomy including the thickness of the epidermis, cortex, vascular tissue, and did not affect the density of glucomannan idioblasts in tubers.
