Abstract
Aquilaria malaccensis merupakan suatu tumbuhan yang memiliki nilai ekonomis tinggi namun saat ini terancam populasi dikarenakan kerusakan habitat. Salah satu kerusakan habitat dialami A. malaccensis adalah pH yang rendah sehingga menimbulkan efek toksisitas alumunium. Dengan ini diperlukan cara untuk bisa meningkatkan populasi dari A. malaccensis yang bisa mentoleransi perubahan pada habitat aslinya. Cara untuk bisa mengatasi masalah ini adalah melalui seleksi in vitro dengan tahapan penelitian yang terdiri dari menumbuhkan eksplan tunas selama 1 bulan dengan media pertumbuhan ½ MS dengan penambahan agar, sukrosa serta variasi komposisi ZPT IBA, BAP, dan mT. Cekaman asam dengan aluminium diseleksi menggunakan media pertumbuhan terbaik, BAP 2 ppm, yang ditambahkan dengan larutan AlCl3. Eksplan diinkubasi pada suhu 20 0C dengan pencahayaan 2000 lux dan fotoperiode 16/8 terang/gelap selama 2 minggu. Hasil penelitian menunjukkan bahwa tinggi, jumlah daun, dan jumlah tunas yang tidak berbeda signifikan terhadap kontrol pada perlakuan konsentrasi AlCl3 25 ppm, 50 ppm dan 75 ppm. Planlet hasil penelitian ini selanjutkan dipakai dalam ekstraksi DNA yang dilanjutkan dengan PCR untuk mendeteksi gen tarkait ketahanan asam yaitu Almt1 dan STOP1. Gen Almt1 dan STOP1 terdeteksi pada 500 bp (Almt1) dan 800 bp (STOP1).

Aquilaria malaccensis is a plant species with high economic value; however, its population is currently threatened due to habitat degradation. One of the major environmental stresses affecting A. malaccensis is low soil pH, which leads to aluminum toxicity. Therefore, an approach is needed to increase the population of A. malaccensis capable of tolerating changes in its natural habitat. One potential strategy is in vitro selection. This study consisted of culturing shoot explants for one month on half-strength ½ MS medium supplemented with agar, sucrose, and various combinations of the plant growth regulators IBA, BAP, and mT. Acid stress induced by aluminum was applied using the best growth medium, containing 2 ppm BAP, supplemented with AlCl₃ solution. The explants were incubated at 20°C under a light intensity of 2,000 lux with a 16/8 h light/dark photoperiod for two weeks. The results showed that plant height, number of leaves, and number of shoots in explants treated with 25, 50, and 75 ppm AlCl₃ were not significantly different from the control. The regenerated plantlets obtained from this study were subsequently used for DNA extraction followed by PCR analysis to detect acid tolerance-related genes, namely Almt1 and STOP1. The Almt1 and STOP1 genes were successfully detected at approximately 500 bp and 800 bp, respectively.
