Abstract

Perkembangan Antimicrobial Resistance (AMR) pada bakteri telah menurunkan efektivitas berbagai antibiotik dalam menghambat pertumbuhan ataupun membunuh bakteri patogen. Salah satu pendekatan yang dikembangkan untuk mengatasi permasalahan tersebut adalah modifikasi antibiotik menggunakan logam tembaga (Cu), yang diketahui memiliki aktivitas antibakteri serta mampu meningkatkan efektivitas senyawa lain melalui interaksi sinergis. Penelitian ini bertujuan untuk meningkatkan aktivitas antibakteri pada ofloksasin melalui dua pendekatan, yaitu interaksi Cu2O-ofloksasin dan senyawa kompleks Cu(ofloksasin)2. Cu2O disintesis melalui reaksi reduksi menggunakan glukosa sebagai pereduksi dan trisodium sitrat (TSC) sebagai agen penstabil. Larutan kemudian dikondisikan pada pH 9, dilanjutkan dengan proses pengadukan hingga didapatkan endapan Cu2O. Cu2O hasil sintesis dicampurkan dengan ofloksasin melalui pengadukan dengan magnetic stirer. Sedangkan, kompleks Cu(ofloksasin)2 berhasil disintesis melalui metode refluks menggunakan rasio molaritas logam dan ligan (1:2) serta mengkondisikan larutan pada pH 7,05. Ketiga senyawa hasil sintesis dikarakterisasi dengan spektrofotometer UV-Vis, FTIR, dan XRD. Ketiga senyawa juga diuji aktivitas antibakterinya terhadap B. subtilis dan P. aeruginosa menggunakan metode difusi cakram. Terbentuknya Cu₂O dikonfirmasi melalui puncak serapan UV-Vis pada 286 nm, pita vibrasi Cu(I)–O pada 628 cm⁻¹ (FTIR), serta puncak difraksi 2θ yang sesuai dengan pola referensi Cu₂O. Karakterisasi Cu2O-ofloksasin mengindikasikan bahwa terdapat molekul ofloksasin yang teradsorpsi pada permukaan Cu2O. Serta kemungkinan mekanisme fisisorpsi melalui interaksi elektrostatik lemah dan ikatan hidrogen. Hasil karakterisasi senyawa kompleks menunjukkan proses kompleksasi melibatkan koordinasi gugus karboksilat ofloksasin terhadap ion Cu²⁺. Δν(COO⁻) sebesar 154 cm⁻¹ pada spektrum FTIR mengindikasikan mode koordinasi bidentat. Pita serapan lebar pada sekitar 650-750 nm pada spektrum UV-Vis dikatikan dengan struktur geometri piramida persegi terdistorsi. Hasil pengujian menunjukkan bahwa peningkatan aktivitas antibakteri hanya terjadi pada kompleks Cu(ofloksasin)2 dengan diameter zona inhibisi 20,40±0,40 mm terhadap bakteri B. subtilis. Namun kedua pendekatan tidak menunjukkan peningkatan aktivitas antibakteri terhadap bakteri P. aeruginosa . Hasil penelitian ini menunjukkan bahwa Cu(ofloksasin)2 lebih efektif meningkatkan aktivitas antibakteri dibandingkan interaksi Cu2O–ofloksasin. ==============================================================================================================================
The development of Antimicrobial Resistance (AMR) in bacteria has made various antibiotics less effective at stopping the growth of or killing disease-causing bacteria. One of the ways developed to tackle this problem is by modifying antibiotics with copper (Cu), which is known to have antibacterial activity and can increase the effectiveness of other compounds through synergistic interactions. This study aims to increase the antibacterial activity of the antibiotic ofloxacin through two approaches: Cu2O-ofloxacin interaction and the Cu(ofloxacin)2 complex compound. Cu2O was synthesized through a reduction reaction using glucose as a reducing agent and trisodium citrate (TSC) as a stabilizing agent, then conditioned at pH 9, and continued with stirring until Cu2O precipitate formed. The synthesized Cu2O was mixed with ofloxacin by stirring with a magnetic stirrer. Meanwhile, the Cu(ofloxacin)₂ complex was successfully synthesized using a reflux method with a metal-to-ligand molar ratio of 1:2 and by conditioning the solution at pH 7.05. The three synthesized compounds were characterized using UV-Vis spectrophotometry, FTIR, and XRD. The antibacterial activity of the three compounds was evaluated against B. subtilis and P. aeruginosa using the disk diffusion method. The formation of Cu₂O was confirmed by a UV-Vis absorption peak at 286 nm, a Cu(I)–O vibrational band at 628 cm⁻¹ (FTIR), and a 2θ diffraction peak consistent with the Cu₂O reference pattern. The results of the characterization of Cu₂O-ofloxacin indicate that adsorption occurs via a physisorption mechanism involving the formation of hydrogen bonds and weak electrostatic interactions. The characterization of the complex compounds shows that the complexation process involves the coordination of the ofloxacin carboxyl group to the Cu²⁺ ion. The Δν(COO⁻) value of 154 cm⁻¹ in the FTIR spectrum indicates a possible bidentate coordination mode. The broad absorption band around 650-750 nm appearing in the UV-Vis spectrum is associated with a distorted square-pyramidal geometry. The test results showed that an increase in antibacterial activity only occurred in the Cu(ofloxacin)2 complex, with an inhibition zone diameter of 20.40±0.40 mm against the Gram-positive bacteria B. subtilis. However, both approaches did not show increased antibacterial activity against Gram-negative bacteria. These results indicate that the Cu(ofloxacin)2 complex is more effective at enhancing antibacterial activity compared to the Cu₂O–ofloxacin interaction.
