Abstract
Padi merupakan sumber pangan utama bagi lebih dari setengah penduduk dunia. Namun, hasil panen padi sangat dipengaruhi oleh cekaman salinitas karena padi merupakan tanaman serealia yang sangat sensitif terhadap kadar garam tinggi. Sebanyak 20% dari 230 juta hektar lahan pertanian di seluruh dunia terkena dampak salinitas. Pengembangan varietas padi yang toleran terhadap cekaman salinitas dilakukan agar lahan salin dapat dimanfaatkan dengan baik. Penelitian ini melakukan pengujian terhadap 5 varietas baru, yaitu varietas ITS 1, ITS2, ITS3, ITS4 dan ITS 5 terhadap cekaman salinitas tinggi. Penelitian ini bertujuan untuk mengamati respon morfologi dan profil transkiptom kelima varietas baru tersebut terhadap cekaman salinitas tinggi. Metode kerja yang dilakukan meliputi pemilihan benih, pembuatan media tanam, penyemaian benih, penanaman, perlakuan salinitas, pemeliharaan tanaman, pengamatan morfologi dan Fisiologi. Ekspresi gen yang dilakukan dengan menggunakan qPCR. Kemudian data dianalisis berdasarkan Rancangan Acak Lengkap menggunakan T-Test. Hasil penelitian ini menunjukkan bahwa cekaman salinitas 100 mM memengaruhi morfologi, fisiologi, dan respon molekuler kelima varietas padi baru dengan respon yang berbeda pada setiap varietasnya. ITS5 menunjukkan respon yang paling konsisten terhadap cekaman salinitas diantara varietas lainnya, ITS5 mampu menunjukkan respon morfologi baik, aktivitas antioksidan yang tinggi, akumulasi prolin yang baik, mempertahankan stabilitas membran sel, serta meningkatkan ekspresi gen OsNAC6 dan OsCATB sebagai mekanisme adaptasi molekuler terhadap cekaman salinitas.

Rice is the primary food source for more than half the world's population. However, rice yields are significantly affected by salinity stress because rice is a cereal crop that is highly sensitive to high salt levels. As much as 20% of the 230 million hectares of agricultural land worldwide are affected by salinity. The development of rice varieties tolerant to salinity stress is carried out to ensure optimal utilization of saline land. This study tested five new varieties, namely ITS 1, ITS2, ITS3, ITS4, and ITS 5, against high salinity stress. This study aimed to observe the morphological responses and transcriptome profiles of these five new varieties to high salinity stress. The work methods used included seed selection, planting media preparation, seed sowing, planting, salinity treatment, plant maintenance, Morphological and physiological observations. Gene expression was performed using qPCR. The data obtained were then analyzed using a completely randomized design using T-Test. The results of this study indicate that 100 mM salinity stress affects the morphology, physiology, and molecular responses of five new rice varieties with different responses in each variety. ITS5 showed the most consistent response to salinity stress among other varieties, ITS5 was able to show good morphological responses, high antioxidant activity, good proline accumulation, maintaining cell membrane stability, and increasing the expression of OsNAC6 and OsCATB genes as a molecular adaptation mechanism to salinity stress.
