Abstract
β-karoten merupakan pigmen karotenoid yang memiliki aktivitas antioksidan tinggi dan berpotensi dimanfaatkan pada industri pangan, farmasi, serta kosmetik. Produksi β-karoten secara biologis menggunakan Neurospora sp. melalui sistem solid-state fermentation (SSF) menjadi salah satu alternatif yang ramah lingkungan karena memanfaatkan limbah organik sebagai substrat fermentasi. Penelitian ini bertujuan mengetahui pengaruh kombinasi substrat limbah organik terhadap produksi β-karoten oleh Neurospora sp. melalui metode solid-state fermentation. Produksi β-karoten dilakukan menggunakan sistem SSF dengan empat substrat tunggal, yaitu tandan kosong kelapa sawit (TKKS), mesokarp dan eksokarp kelapa sawit, kulit buah kopi, dan ampas tahu, serta tujuh kombinasi substrat. Kadar β-karoten dianalisis menggunakan spektrofotometer UV-Vis pada panjang gelombang 450 nm dan dinyatakan sebagai yield β-karoten (µg/g biomassa). Hasil penelitian menunjukkan bahwa kombinasi substrat limbah organik memberikan pengaruh terhadap produksi β-karoten oleh Neurospora sp. Kombinasi kulit buah kopi dan ampas tahu (SK5) menghasilkan yield β-karoten tertinggi, yaitu sebesar 67,99 ± 8,51 µg/g biomassa, sedangkan substrat yang mengandung tandan kosong kelapa sawit (TKKS) cenderung menghasilkan yield lebih rendah, yaitu berkisar antara 3,85 ± 1,09 hingga 27,86 ± 5,14 µg/g biomassa. Dengan demikian, kombinasi substrat limbah organik yang memiliki komposisi nutrisi saling melengkapi, terutama keseimbangan sumber karbon dan nitrogen lebih efektif dalam meningkatkan produksi β-karoten oleh Neurospora sp. melalui sistem SSF.

β-Carotene is a carotenoid pigment with high antioxidant activity and has broad potential applications in the food, pharmaceutical, and cosmetic industries. The biological production of β-carotene using Neurospora sp. through a solid-state fermentation (SSF) system represents an environmentally friendly alternative by utilizing organic waste as fermentation substrates. This study aimed to determine the effect of different combinations of organic waste substrates on β-carotene production by Neurospora sp. using the solid-state fermentation method. β-Carotene production was carried out using the SSF system with four single substrates, namely oil palm empty fruit bunches (OPEFB), oil palm mesocarp and exocarp, coffee husk, and tofu pulp (okara), as well as seven substrate combinations. The β-carotene content was determined using a UV–Vis spectrophotometer at a wavelength of 450 nm and expressed as β-carotene yield (µg/g biomass). The combination of coffee pulp and tofu residue (SK5) produced the highest β-carotene yield, reaching 67.99 ± 8.51 µg/g biomass, whereas substrates containing oil palm empty fruit bunches (OPEFB) generally resulted in lower yields, ranging from 3.85 ± 1.09 to 27.86 ± 5.14 µg/g biomass. Therefore, combinations of organic waste substrates with complementary nutrient compositions, particularly a balanced supply of carbon and nitrogen sources, were more effective in enhancing β-carotene production by Neurospora sp. through the SSF system.
