Abstract
Produksi β-karoten alami semakin banyak dibutuhkan saat ini karena manfaatnya yang luas dalam sektor pangan, Kesehatan, kosmetik, dan industri. Neurospora sp. diketahui dapat tumbuh dengan cepat pada substrat lignoselulosa dan dapat menghasilkan pigmen β-karoten. Pemanfaatan limbah padat kelapa sawit (TKKS dan sisa buah sawit) sebagai substrat solid-state fermentation (SSF) akan memberikan peluang berupa pengelolaan limbah berkelanjutan serta produksi pigmen yang bernilai tambah. Penelitian ini bertujuan untuk mengetahui pengaruh suhu inkubasi (suhu ruang dan 35°C) dan pH media fermentasi (4,5, dan 6) terhadap produksi β-karoten menggunakan pendekatan SSF selama tujuh hari. Produksi β-Karoten tertinggi diperoleh pada kombinasi suhu ruang dan pH 5 sebesar 36,883 ppm, sedangkan produksi terendah diperoleh pada kombinasi suhu 35°C dan pH 5 sebesar 4,878 ppm. Hasil penelitian ini menunukkan bahwa kombinasi suhu ruang dan pH awal 5 merupakan kondisi terbaik dalam rentang perlakuan yang diuji untuk produksi β-Karoten pada Neurospora sp. Selain itu, hasil ini menunjukkan bahwa faktor lingkungan dan interaksinya berperan dalam menentukan produksi β-Karoten, serta menunjukkan adanya potensi limbah padat kelapa sawit sebagai substrat alternatif untuk produksi β-Karoten melalui SSF.

Natural β-carotene production is increasingly in demand due to its wide range of applications in the food, health, cosmetic, and industrial sectors. Neurospora sp. is known for its ability to grow rapidly on lignocellulosic substrates and produce β-carotene pigments. The utilization of palm oil solid waste, including oil palm empty fruit bunches (OPEFB) and oil palm fruit residues, as substrates for solid-state fermentation (SSF) offers opportunities for sustainable waste management and the production of value-added pigments. This study aimed to determine the effects of incubation temperature (room temperature and 35°C) and fermentation medium pH (4, 5, and 6) on β-carotene production using an SSF approach for seven days. The highest β-carotene production was obtained at the combination of room temperature and pH 5, reaching 36,883 ppm, whereas the lowest production was observed at 35°C and pH 5, with a concentration of 4,878 ppm. These findings indicate that the combination of room temperature and an initial pH of 5 represents the most favorable condition within the tested treatment range for β-carotene production by Neurospora sp. Furthermore, the results suggest that environmental factors and their interactions play a role in determining β-carotene production and demonstrate the potential of palm oil solid waste as an alternative substrate for β-carotene production through SSF.
