Abstract
Peningkatan konsumsi produk pangan olahan hewani yang praktis dan ekonomis mendorong adanya jaminan kehalalan dan keamanan pangan, khususnya pada produk yang berpotensi mengalami kecurangan komposisi daging. Pencampuran daging halal dan non-halal pada produk olahan seperti bakso, sosis, dan nugget dapat merugikan konsumen serta menimbulkan risiko kesehatan. Metode identifikasi berbasis morfologi dan protein memiliki keterbatasan pada produk yang telah mengalami proses pemanasan dan penggilingan. Oleh karena itu, penelitian ini bertujuan menguji validitas primer multiplex polymerase chain reaction (mPCR) dalam mengidentifikasi kandungan spesies daging pada produk olahan hewani. Penelitian dilakukan menggunakan produk olahan dari daging sapi (Bos taurus), kambing (Capra hircus), ayam (Gallus gallus domesticus), tikus (Rattus norvegicus), dan babi (Sus scrofa), tanpa domba (Ovis aries) dan kuda (Equus caballus). Tahapan penelitian terdiri atas ekstraksi DNA, pengukuran konsentrasi DNA menggunakan Quantus fluorometer, amplifikasi DNA dengan metode mPCR, dan analisis hasil amplifikasi menggunakan elektroforesis gel agarosa. Hasil penelitian menunjukkan bahwa primer mPCR yang digunakan memiliki validitas yang baik dalam mengidentifikasi spesies daging pada produk olahan hewani. 27 dari 30 sampel menunjukkan pita DNA spesifik yang sesuai dengan komposisi spesies pada produk, sehingga diperoleh tingkat validitas sebesar 90%. Dengan demikian, primer multiplex PCR yang digunakan valid untuk mengidentifikasi kandungan spesies daging pada produk olahan hewani.

The increasing consumption of practical and affordable processed meat products has heightened the need for halal assurance and food safety, particularly in products that are vulnerable to meat adulteration. The incorporation of halal and non-halal meat in processed products such as meatballs, sausages, and nuggets may mislead consumers and pose potential health risks. Conventional identification methods based on morphological and protein characteristics have limitations when applied to products that have undergone extensive processing, including heating and grinding. Therefore, this study aimed to evaluate the validity of multiplex polymerase chain reaction (mPCR) primers for identifying meat species in processed animal-based food products. The study was conducted using processed products derived from beef (Bos taurus), goat (Capra hircus), chicken (Gallus gallus domesticus), rat (Rattus norvegicus), and pork (Sus scrofa), excluding sheep (Ovis aries) and horse (Equus caballus). The research procedures included DNA extraction, measurement of DNA concentration using a Quantus fluorometer, DNA amplification using the mPCR method, and analysis of amplification products through agarose gel electrophoresis. The results demonstrated that the mPCR primers used exhibited good validity for identifying meat species in processed animal-based products. A total of 27 out of 30 samples produced specific DNA bands corresponding to the declared species composition, resulting in a validity rate of 90%. These findings indicate that the multiplex PCR primers used are valid for identifying meat species content in processed animal-based products.
