Abstract

Ganoderma boninense merupakan patogen tular tanah yang sangat virulen penyebab penyakit busuk pangkal batang kelapa sawit dan mampu bertahan lama di tanah maupun jaringan tanaman. Perubahan komunitas mikroba di rizosfer akibat infeksi patogen ini membuka peluang pemanfaatan bakteri rizosfer adaptif sebagai agen pengendali hayati. Penelitian ini bertujuan mengevaluasi kemampuan isolat bakteri rizosfer dalam menghambat pertumbuhan G. boninense secara tunggal maupun dalam bentuk konsorsium dua dan tiga isolat serta mengidentifikasi aktivitas enzim selulase, ligninase, kitinase, dan siderofor sebagai mekanisme pendukung antagonisme. Metode yang digunakan meliputi pengujian antagonisme secara in vitro menggunakan isolat tunggal, konsorsium cair, dan konsorsium padat terhadap G.boninense, pengukuran Percentage Inhibition of Radial Growth (PIRG), serta uji kualitatif enzim selulase, ligninase, kitinase, dan siderofor. Hasil uji antagonisme menunjukkan bahwa isolat tunggal 2N4, 0N5, 2S1, dan 2S2 memiliki kemampuan penghambatan tertinggi terhadap G. boninense dengan nilai PIRG berturut-turut sebesar 27,35%; 23,04%; 20,89%; dan 20,08%. Pada perlakuan konsorsium cair, kombinasi 2S1–0N5 menunjukkan aktivitas antagonis tertinggi dengan nilai PIRG sebesar 21,16%, sedangkan pada konsorsium padat, kombinasi 0N5–2N4–2S2 menghasilkan nilai PIRG tertinggi sebesar 12,43%. Hasil analisis One-Way ANOVA yang dilanjutkan dengan uji post hoc Tukey menunjukkan bahwa perlakuan isolat tunggal berbeda nyata dibandingkan perlakuan konsorsium cair dan konsorsium padat. Hasil uji kualitatif menunjukkan bahwa isolat 2N4, 0N5, 2S1, dan 2S2 mampu menghasilkan enzim selulase, kitinase, dan siderofor, sedangkan aktivitas ligninase hanya terdeteksi pada isolat 2N4 dan 2S1. Hasil penelitian ini menunjukkan bahwa bakteri rizosfer memiliki potensi sebagai agen pengendali hayati terhadap G. boninense , dengan perlakuan bakteri tunggal menunjukkan kemampuan antagonis yang lebih baik dibandingkan perlakuan konsorsium.

Ganoderma boninense is a highly virulent soil-borne pathogen that causes basal stem rot disease in oil palm and is capable of surviving for extended periods in soil and plant tissues. Changes in the rhizosphere microbial community resulting from pathogen infection provide opportunities for utilizing adaptive rhizosphere bacteria as biological control agents. This study aimed to evaluate the ability of rhizosphere bacterial isolates to inhibit the growth of G. boninense individually and in two- and three-isolate consortia, as well as to identify cellulase, ligninase, chitinase, and siderophore activities as supporting mechanisms of antagonism. The methods included in vitro antagonism assays using single isolates, liquid consortia, and solid consortia against G. boninense , measurement of Percentage Inhibition of Radial Growth (PIRG), and qualitative assays for cellulase, ligninase, chitinase, and siderophore production. The antagonism assay results showed that single isolates 2N4, 0N5, 2S1, and 2S2 exhibited the highest inhibitory activity against G. boninense , with PIRG values of 27.35%, 23.04%, 20.89%, and 20.08%, respectively. In the liquid consortium treatment, the combination of 2S1–0N5 showed the highest antagonistic activity with a PIRG value of 21.16%, whereas in the solid consortium treatment, the combination of 0N5–2N4–2S2 produced the highest PIRG value of 12.43%. One-Way ANOVA followed by Tukey’s post hoc test revealed that the single-isolate treatment differed significantly from both the liquid and solid consortium treatments. Qualitative assays indicated that isolates 2N4, 0N5, 2S1, and 2S2 were capable of producing cellulase, chitinase, and siderophores, while ligninase activity was detected only in isolates 2N4 and 2S1. These findings demonstrate that rhizosphere bacteria have potential as biological control agents against G. boninense , with single-isolate treatments exhibiting greater antagonistic activity than consortium treatments.
