Abstract

Kerusakan daun akibat aktivitas serangga herbivor dapat memengaruhi kemampuan fotosintesis, pertumbuhan, dan kondisi fisiologis tanaman mangrove. Penelitian ini bertujuan menganalisis tingkat kerusakan daun Avicennia marina akibat serangan serangga herbivor menggunakan metode Leaf Damage Index (LDI), mengetahui kandungan kimia nutrien daun (nitrogen, serat kasar, flavonoid, tanin, alkaloid, kadar air, dan polifenol), serta menganalisis pola profil kandungan kimiawi tersebut terhadap tingkat herbivori pada daun muda dan daun tua. Penelitian dilaksanakan di Kebun Raya Mangrove Gunung Anyar, Surabaya, pada bulan April–Mei 2026. Pengukuran luas daun dilakukan secara non-destruktif menggunakan ImageJ untuk menghitung persentase herbivori yang selanjutnya diklasifikasikan berdasarkan LDI, sementara kandungan kimia daun dianalisis di Laboratorium LITHR, Universitas Airlangga. Hasil penelitian menunjukkan bahwa daun muda mengalami kerusakan lebih tinggi dibandingkan daun tua, dengan rata-rata luas daun rusak sebesar 32,41 mm² (kategori V/Tinggi; 26,70%) berbanding 2.114 mm² pada daun tua (kategori IV/Sedang; 12,31%). Daun muda memiliki kadar air (74,8%) dan nitrogen (0,73%) lebih tinggi dibandingkan daun tua (67,0%; 0,70%), sedangkan daun tua memiliki kandungan tanin, polifenol, alkaloid, dan serat kasar yang lebih tinggi; saponin justru lebih tinggi pada daun muda. Hasil ini menunjukkan bahwa daun muda lebih rentan terhadap herbivori karena kandungan nutrien tinggi namun pertahanan kimia belum berkembang optimal, sedangkan daun tua memiliki pertahanan kimia lebih kuat melalui akumulasi metabolit sekunder sehingga kerusakannya lebih rendah

Leaf damage caused by herbivorous insects can affect the photosynthetic capacity, growth, and physiological condition of mangrove plants. This study aimed to analyze the level of leaf damage in Avicennia marina caused by herbivorous insects using the Leaf Damage Index (LDI) method, to determine the nutrient and chemical content of the leaves (nitrogen, crude fiber, flavonoids, tannins, alkaloids, water content, and phenolics), and to analyze the profile pattern of these chemical compounds in relation to the herbivory level of young and mature leaves. The research was conducted at the Gunung Anyar Mangrove Botanical Garden, Surabaya, from April to May 2026. Leaf area was measured non-destructively using ImageJ software to calculate the herbivory percentage, which was subsequently classified according to the LDI, while the leaf chemical content was analyzed at the LITHR Laboratory, Universitas Airlangga. The results showed that young leaves experienced greater damage than mature leaves, with an average damaged leaf area of 32,41 mm² (Category V/High; 26,70%) compared to 2,114 mm² in mature leaves (Category IV/Moderate; 12,31%). Young leaves had higher water content (74.8%) and nitrogen (0.73%) than mature leaves (67.0%; 0.70%), whereas mature leaves showed higher concentrations of tannins, polyphenols, alkaloids, and crude fiber; saponin content, however, was higher in young leaves. These findings indicate that young leaves are more susceptible to herbivory due to their higher nutritional value but less-developed chemical defenses, whereas mature leaves possess stronger chemical defenses through the accumulation of secondary metabolites, resulting in lower levels of herbivore damage.
