Abstract
Kolagen merupakan biomaterial yang banyak dimanfaatkan di bidang medis, pangan, kosmetik, dan bioteknologi. Limbah ikan tuna diketahui mengandung kolagen yang tinggi sehingga limbah ikan tuna dapat digunakan sebagai sumber kolagen dengan skrining bakteri pendegradasi kolagen. Penelitian ini bertujuan untuk mengisolasi dan mengkarakterisasi fenotip bakteri dari limbah ikan tuna, dan mengetahui hubungan kekerabatannya menggunakan pendekatan taksonomi numerik fenetik. Isolasi dilakukan pada media NA yang diperkaya kolagen dan diperoleh 14 isolat bakteri. Berdasarkan pembentukan zona bening dipilih lima isolat, yaitu LIT 1, LIT 4, LIT 5, LIT 10, dan LIT 11, untuk dikarakterisasi lebih lanjut meliputi morfologi koloni, bentuk sel, pewarnaan Gram, pembentukan endospora, uji biokimia, dan uji fisiologi. Data karakter fenotip ditabulasikan ke dalam tabel n x t dan konstruksi dendogram menggunakan aplikasi Microsoft Excel dan program MVSP (Multivariate Statistical Package) versi 3.22.. Hasil analisis menunjukkan nilai similaritas antar isolat berkisar antara 54,8 - 90,3% dan membentuk tiga cluster, yaitu cluster A (LIT 1), cluster B (LIT 4), dan cluster C (LIT 5, LIT 10, dan LIT 11). Analisis korelasi kofenetik menghasilkan R² sebesar 0,8526 dengan R sebesar 0,92, yang menunjukkan bahwa dendrogram yang dihasilkan telah merepresentasikan matriks similaritas dengan baik. Berdasarkan Generic Assignment dengan metode profile matching menggunakan panduan dari Bergey's manual of determinative bacteriology teridentifikasi adalah Enterobacter (LIT 1), Bacillus (LIT 4), dan Neisseria (LIT 5, LIT 10, dan LIT 11).

Collagen is a biomaterial widely utilized in the medical, food, cosmetic, and biotechnology sectors. Tuna waste is known to have a high collagen content, making it a potential source of collagen that can be screened for collagen-degrading bacteria. This study aimed to isolate and characterize the phenotypes of bacteria from tuna waste and to determine their phylogenetic relationships using a phenetic numerical taxonomy approach. Isolation was performed on collagen-enriched Nutrient Agar (NA) medium, yielding 14 bacterial isolates. Based on the formation of clear zones, five isolates LIT 1, LIT 4, LIT 5, LIT 10, and LIT 11 were selected for further characterization, including colony morphology, cell shape, Gram staining, endospore formation, and biochemical and physiological tests. Phenotypic character data were tabulated into an n x t matrix, and a dendrogram was constructed using Microsoft Excel and the MVSP (Multivariate Statistical Package) program version 3.22. Analysis results showed similarity values between isolates ranging from 54.8% to 90.3%, forming three clusters: cluster A (LIT 1), cluster B (LIT 4), and cluster C (LIT 5, LIT 10, and LIT 11). Cophenetic correlation analysis yielded an R² of 0.8526 and an R of 0.92, indicating that the resulting dendrogram accurately represented the similarity matrix. Based on generic assignment using the profile matching method and Bergey's Manual of Determinative Bacteriology as a reference, the isolates were identified as Enterobacter (LIT 1), Bacillus (LIT 4), and Neisseria (LIT 5, LIT 10, and LIT 11).
