Abstract
Resin kemenyan (Styrax spp.) dari Sumatera Utara merupakan sumber alami yang kaya akan senyawa metabolit sekunder, salah satunya asam benzoat yang memiliki nilai strategis tinggi sebagai bahan baku industri pangan, farmasi, dan kosmetik. Penelitian ini bertujuan untuk memetakan profil metabolit 14 sampel ekstrak etanol resin Styrax spp., dari empat lokasi di Sumatera Utara dengan variasi ketinggian tempat tumbuh menggunakan GC–MS dan LC–MS, mengisolasi asam benzoat dari sampel terpilih, serta sintesis natrium benzoat dan benzil benzoat dari isolat yang diperoleh. Isolasi asam benzoat dari 20,23 gram ekstrak melalui saponifikasi dengan KOH 2M, pengasaman dengan H2SO4 25%, dan rekristalisasi menghasilkan 0,61 gram isolat dengan rendemen 3,07%. Isolat berupa padatan putih menyerupai salju halus dengan titik leleh 111 °C. Kemurnian dikonfirmasi melalui KLT tiga sistem eluen dan dua dimensi yang menunjukkan bercak tunggal, serta ¹H-NMR dalam DMSO-d₆ yang mengkonfirmasi identitas asam benzoat. Sintesis natrium benzoat melalui reaksi netralisasi menghasilkan 0,21 gram produk dengan rendemen 75,29%, dikonfirmasi melalui ¹H-NMR dalam D2O. Sintesis benzil benzoat melalui reaksi substitusi nukleofilik bimolekular menggunakan benzil klorida dalam DMF menghasilkan 0,37 gram produk dengan rendemen 88,00%, dikonfirmasi melalui ¹H-NMR dalam CDCl3 dengan sinyal penciri –OCH₂– pada δ 5,297–5,381 ppm. Penelitian ini membuktikan bahwa resin kemenyan Sumatera Utara dapat dimanfaatkan sebagai sumber asam benzoat untuk sintesis senyawa turunan bernilai industri tinggi.

Benzoin resin (Styrax spp.) from North Sumatra is a rich natural source of secondary metabolites, particularly benzoic acid, which holds high strategic value as a raw material in the food, pharmaceutical, and cosmetic industries. This study aims to map the metabolite profiles of 14 ethanol extract samples of Styrax spp., resin from four locations in North Sumatra across various growing altitudes using GC–MS and LC–MS, isolate benzoic acid from the selected sample, and synthesize sodium benzoate and benzyl benzoate from the obtained isolate. The isolation of benzoic acid from 20.23 grams of extract via saponification with 2M KOH, acidification with 25% H2SO4, and recrystallization yielded 0.61 grams of isolate with a 3.07% yield. The isolate was obtained as a fine, snow-like white solid with a melting point of 111°C. Purity was confirmed through thin-layer chromatography (TLC) using three eluent systems and two dimensions, which revealed a single spot, as well as 1H-NMR in DMSO-d6 that confirmed the identity of benzoic acid. The synthesis of sodium benzoate via a neutralization reaction produced 0.21 grams of product with a 75.29% yield, confirmed by 1H-NMR in D2O. The synthesis of benzyl benzoate via an bimolacular nucleophilic substitution reaction using benzyl chloride in DMF yielded 0.37 grams of product with an 88.00% yield, confirmed by 1H-NMR in CDCl3 showing a characteristic –OCH2– signal at δ 5.297–5.381 ppm. This study demonstrates that North Sumatran benzoin resin can be effectively utilized as a natural source of benzoic acid for the synthesis of high-value industrial derivative compounds.
