Abstract
Metil Oranye (MO) merupakan senyawa golongan pewarna azo yang banyak digunakan dalam bidang industri tekstil. Pada penelitian ini digunakan metode biodegradasi pewarna MO oleh bakteri Bacillus subtilis yang terimobilisasi dalam matriks SA-PVA-Bentonit pada kondisi optimum yakni 36,6℃, pH 6,4, selama 6 hari waktu inkubasi. Hasil dari proses biodegradasi pewarna MO dikarakterisasi menggunakan instrumen LC-MS untuk mengidentifikasi senyawa metabolit hasil biodegradasi MO. Identifikasi produk senyawa metabolit dari proses degradasi MO menghasilkan senyawa antara lain p-hidroksi-dimetilamino azobenzene (RT: 1,15 m/z 256), 4,1–((dihidroksimetil)hyrazono-4) 5-benzena sulfonat (RT: 1,6 m/z 235), 4-Aminobenzenesulfonic acid (RT: 1,9 m/z 254), 4-(2-metilbut-2-en-1-il) diazenil) benzenasulfonat (RT: 2,1 m/z 179), 4-[(2-hydroxy-1-methylpropyl)hydrazono]benzenasulfonat (RT: 2,49 m/z 251), (2-(4-Aminophenyl)hydrazinyl)methanediol (RT: 2,7 m/z 169).

Methyl Orange (MO) is an azo dye compound widely used in the textile industry. In this study, the biodegradation of MO dye by Bacillus subtilis bacterium immobilized in an SA-PVA-Bentonite matrix was studied under optimal conditions of 36.6°C, pH 6.4, for a 6-day incubation period. The results of MO dye biodegradation process were characterized using LC-MS instruments to identify the MO degradation metabolites. The identification of metabolite compounds from the MO degradation process produced compounds including p-hydroxy-dimethylamino azobenzene (RT: 1.15 m/z 256), 4,1–((dihydroxymethyl)hyrazono-4) 5-benzenesulfonate (RT: 1.6 m/z 235), 4-aminobenzenesulfonic acid (RT: 1.9 m/z 254), 4-(2-methylbut-2-en-1-yl) diazenyl) benzenesulfonate (RT: 2.1 m/z 179), 4-[(2-hydroxy-1-methylpropyl)hydrazono] benzenesulfonate (RT: 2.49 m/z 251), (2-(4-Aminophenyl)hydrazinyl)methanediol (RT: 2.7 m/z 169).
