Abstract
Ampas tebu merupakan limbah lignoselulosa yang berpotensi dimanfaatkan sebagai sumber arabinoksilan untuk produksi xilooligosakarida (XOS). Penelitian ini bertujuan untuk mengekstraksi arabinoksilan dari ampas tebu, menghidrolisisnya secara enzimatik menggunakan xilanase, serta mengevaluasi potensinya sebagai kandidat prebiotik fungsional pada bakteri Lactobacillus dan yoghurt sinbiotik. Ampas tebu terlebih dahulu dipretreatment melalui pencucian, pengeringan, pemisahan bagian dinding, penghalusan, dan pengayakan. Hasil karakterisasi awal menunjukkan kadar air ampas tebu sebesar 7,43 ± 0,27% dan kandungan hemiselulosa sebesar 22,68 ± 0,31%. Ekstraksi arabinoksilan dilakukan menggunakan KOH 1%, 3%, dan 5%, dengan hasil terbaik pada KOH 3%, yaitu rendemen 8,60 ± 0,66% sebelum bleaching dan 0,587 ± 0,029% setelah bleaching. Proses bleaching meningkatkan karakteristik arabinoksilan, ditunjukkan oleh perubahan warna menjadi lebih cerah, peningkatan kelarutan dari 35,93 ± 0,47% menjadi 69,41 ± 0,50%, serta penurunan absorbansi pada 420 nm dari 0,382 ± 0,064 menjadi 0,312 ± 0,051. Karakterisasi FTIR menunjukkan adanya gugus khas polisakarida, seperti O–H, C–H, dan C–O/C–O–C glikosidik, serta berkurangnya indikasi komponen aromatik lignin setelah proses bleaching. Hidrolisis enzimatik menghasilkan gula pereduksi, dan analisis KLT serta HPLC menunjukkan adanya indikasi terbentuknya campuran produk hidrolisis, termasuk kemungkinan XOS, terutama pada hidrolisat 4 jam. Hasil uji prebiotik menunjukkan bahwa hidrolisat mampu mendukung pertumbuhan Lactobacillus, dengan jumlah koloni tertinggi pada konsentrasi 3%. Pada aplikasi yoghurt sinbiotik, penambahan hidrolisat arabinoksilan bleaching menghasilkan pH terendah dan total asam tertitrasi tertinggi pada hari ke-6, yaitu pH 3,06 dan TAT 1,085 ± 0,021%. Hasil penelitian ini menunjukkan bahwa hidrolisat arabinoksilan dari ampas tebu berpotensi sebagai kandidat prebiotik fungsional untuk pengembangan yoghurt sinbiotik.

Sugarcane bagasse is a lignocellulosic waste with potential as a source of arabinoxylan to production of Xylo-oligosaccharides (XOS). This study aimed to extract arabinoxylan from sugarcane bagasse, enzymatically hydrolyze it with xylanase, and evaluate its potential as a functional prebiotic for Lactobacillus growth and synbiotic yogurt development. Sugarcane bagasse was pretreated by washing, drying, separating the outer fibrous fraction, grinding, and sieving. Initial characterization showed that the pretreated bagasse had a moisture content of 7.43 ± 0.27% and hemicellulose content of 22.68 ± 0.31%. Arabinoxylan extraction was carried out using 1%, 3%, and 5% KOH, with the best result obtained at 3% KOH, yielding 8.60 ± 0.66% before bleaching and 0.587 ± 0.029% after bleaching. The bleaching process improved the characteristics of arabinoxylan, as indicated by a lighter color, increased solubility from 35.93 ± 0.47% to 69.41 ± 0.50%, and decreased absorbance at 420 nm from 0.382 ± 0.064 to 0.312 ± 0.051. FTIR characterization confirmed the presence of typical polysaccharide functional groups, including O–H, C–H, and glycosidic C–O/C–O–C groups, as well as a reduced indication of aromatic lignin components after bleaching. Enzymatic hydrolysis produced reducing sugars, while TLC and HPLC analyses indicated the formation of mixed hydrolysis products, including possible XOS, particularly in the 4-hour hydrolysate. The preliminary prebiotic assay showed that the hydrolysate supported the growth of Lactobacillus, with the highest colony number observed at a concentration of 3%. In synbiotic yogurt application, the addition of bleached arabinoxylan hydrolysate resulted in the lowest pH and highest titratable acidity on day 6, with a pH of 3.06 and TAT of 1.085 ± 0.021%. These results indicate that arabinoxylan hydrolysate from sugarcane bagasse has potential as a functional prebiotic candidate for synbiotic yogurt development.
