Abstract

Analisis besi (Fe) menggunakan spektrofotometri UV-Vis dengan ligan 1,10-fenantrolin memerlukan agen pereduksi untuk mereduksi Fe3+ menjadi Fe2+. Penelitian ini bertujuan untuk membandingkan kinerja reduktor NH2OH·HCl, gambir (Uncaria gambir), dan natrium oksalat (Na2C2O4), serta mengevaluasi toleransinya terhadap interferensi ion Cu²⁺. Metode penelitian meliputi optimasi kondisi analisis (pH, waktu reaksi, dan konsentrasi pereduksi), pembuatan kurva kalibrasi dan uji interferensi Cu2+ pada rentang konsentrasi 0–0,5 ppm. Hasil penelitian menunjukkan bahwa pH optimum untuk NH2OH·HCl, gambir, dan natrium oksalat berturut-turut adalah 4,5; 3,5; dan 5,0, dengan waktu reaksi optimum masing-masing 15; 90; dan 45 menit. Sensitivitas (slope) tertinggi dimiliki NH₂OH·HCl (0,0962; R2 = 0,9967), diikuti gambir (0,0778; R2 = 0,9980) dan natrium oksalat (0,0258; R2 = 0,9941). Uji interferensi menunjukkan bahwa NH2OH·HCl dan gambir memiliki toleransi yang baik terhadap Cu2+ (mulai mengganggu pada konsentrasi 0,4 ppm), sedangkan natrium oksalat sangat sensitif (mulai mengganggu pada 0,1 ppm). Gambir terbukti menjadi alternatif reduktor alami (green reducing agent) yang sangat menjanjikan dengan performa dan ketahanan terhadap interferensi yang setara dengan NH2OH·HCl.

The analysis of iron (Fe) using UV-Vis spectrophotometry with 1,10-phenanthroline requires a reducing agent to convert Fe³⁺ into Fe²⁺. This study aims to compare the performance of hydroxylamine hydrochloride (NH₂OH·HCl), gambir (*Uncaria gambir*), and sodium oxalate (Na₂C₂O₄) as reducing agents, as well as to evaluate their tolerance toward Cu²⁺ ion interference. The methodology included the optimization of analytical conditions, including pH, reaction time, and reducing agent concentration, followed by the construction of calibration curves and Cu²⁺ interference testing over a concentration range of 0–0.5 ppm. The results showed that the optimum pH values for NH₂OH·HCl, gambir, and sodium oxalate were 4.5, 3.5, and 5.0, respectively, with optimum reaction times of 15, 90, and 45 minutes. NH₂OH·HCl exhibited the highest analytical sensitivity, with a calibration curve slope of 0.0962 (R² = 0.9967), followed by gambir with a slope of 0.0778 (R² = 0.9980), and sodium oxalate with a slope of 0.0258 (R² = 0.9941). Interference testing revealed that NH₂OH·HCl and gambir demonstrated good tolerance toward Cu²⁺ ions, with interference occurring at concentrations of 0.4 ppm and above, whereas sodium oxalate was considerably more sensitive, with interference observed at concentrations as low as 0.1 ppm. Overall, gambir proved to be a promising natural (green) reducing agent, exhibiting analytical performance and resistance to Cu²⁺ interference comparable to those of NH₂OH·HCl.
