Abstract

Curcuma zedoaria (temu putih) merupakan tanaman obat dari famili Zingiberaceae yang diketahui mengandung berbagai metabolit sekunder dan berpotensi sebagai sumber senyawa bioaktif. Penelitian ini bertujuan untuk mengkarakterisasi kandungan metabolit sekunder, menentukan kadar total fenolik (TPC) dan total flavonoid (TFC), mengevaluasi aktivitas antioksidan, mengidentifikasi profil metabolit menggunakan liquid chromatography–mass spectrometry (LC-MS), serta mengevaluasi kualitas DNA genom Curcuma zedoaria asal Nusa Tenggara Timur. Penelitian dilakukan melalui skrining fitokimia, penetapan TPC dan TFC, pengujian aktivitas antioksidan menggunakan metode DPPH dan ABTS, analisis profil metabolit menggunakan LC-MS, serta isolasi dan evaluasi kualitas DNA genom menggunakan metode CTAB, Qubit, dan NanoDrop. Hasil penelitian menunjukkan bahwa ekstrak Curcuma zedoaria mengandung flavonoid, fenolik, terpenoid, steroid, dan tanin. Nilai TPC, TFC, dan aktivitas antioksidan berbeda pada setiap jenis pelarut, menunjukkan bahwa pemilihan pelarut memengaruhi senyawa yang terekstraksi. Analisis LC-MS berhasil mengidentifikasi berbagai metabolit, di antaranya zedoalactone B, tetrahydrocurcumin, hexahydrocurcumin, caryophyllene oxide, dan atractylenolide III. Isolasi DNA genom menghasilkan DNA untai ganda dengan konsentrasi 26,2 ng/µL, rasio A260/A280 sebesar 1,88, dan rasio A260/A230 sebesar 1,86 yang menunjukkan kualitas DNA cukup baik berdasarkan parameter yang diukur. Hasil penelitian ini menunjukkan bahwa Curcuma zedoaria asal Nusa Tenggara Timur mengandung berbagai metabolit sekunder dan berpotensi sebagai sumber senyawa bioaktif serta menyediakan data awal mengenai kualitas DNA genom yang dapat dimanfaatkan pada penelitian molekuler lanjutan.

Curcuma zedoaria (white turmeric) is a medicinal plant belonging to the Zingiberaceae family that contains various secondary metabolites and has potential as a source of bioactive compounds. This study aimed to characterize its secondary metabolite composition, determine the Total Phenolic Content (TPC) and Total Flavonoid Content (TFC), evaluate its antioxidant activity, identify its metabolite profile using liquid chromatography–mass spectrometry (LCMS), and assess the quality of genomic DNA isolated from Curcuma zedoaria collected in East Nusa Tenggara, Indonesia. The study included phytochemical screening, determination of TPC and TFC, antioxidant activity assays using DPPH and ABTS methods, metabolite profiling by LC-MS, and genomic DNA isolation and quality assessment using the CTAB method, Qubit fluorometer, and NanoDrop spectrophotometer. The results showed that the extracts contained flavonoids, phenolics, terpenoids, steroids, and tannins. The TPC, TFC, and antioxidant activities varied among extraction solvents, indicating that solvent selection influenced the composition of extracted compounds. LC-MS analysis identified several metabolites, including zedoalactone B, tetrahydrocurcumin, hexahydrocurcumin, caryophyllene oxide, and atractylenolide III. Genomic DNA isolation yielded double-stranded DNA with a concentration of 26.2 ng/µL, an A260/A280 ratio of 1.88, and an A260/A230 ratio of 1.86, indicating acceptable DNA quality based on the measured parameters. These findings demonstrate that Curcuma zedoaria from East Nusa Tenggara contains diverse secondary metabolites with potential as a source of bioactive compounds and provides preliminary information on genomic DNA quality that may support future molecular studies.
