Abstract

Diabetes mellitus (DM) merupakan penyakit tidak menular yang dijumpai
sebagai diabetes tipe 1 dan tipe 2. DM tipe 2 yang paling umum dijumpai (90-95%)
disebabkan oleh resistensi insulin dan gangguan sekresi insulin. Pengobatan DM
tipe 2 dilakukan dengan mengontrol kadar gula dalam darah dalam bentuk mengatur
pola makan, membatasi jumlah konsumsi karbohidrat, olahraga secara rutin, dan
mengkonsumsi obat penghambat enzim α-glukosidase. Efek samping dari
penggunaan obat tersebut adalah diare, sakit perut, perut kembung, hipoglikemia,
kerusakan hati, resistensi obat, kenaikan berat badan, gangguan pada jantung, serta
biaya yang tinggi. Amida dari asam sinamat menunjukkan potensi sebagai lead
compounds dalam pengembangan senyawa-senyawa penghambat enzim α
glukosidase. Sintesis amida tersebut dilakukan dengan melibatkan tionil klorida
yang termasuk dalam daftar Chemical Weapons Conventions, dan Bahan Kimia
Daftar-3 di Undang-undang Republik Indonesia Nomor 9 tahun 2008 tentang
penggunaan bahan kimia dan larangan penggunaan bahan kimia sebagai senjata
kimia; akibatnya penelitian yang dilakukan berhasil mengembangkan metoda yang
tidak melibatkan tionil klorida. Penelitian yang telah dilakukan berhasil mensintesis
empat amida baru berupa senyawa (E)-N'-(3-fenil)akriloil)isonikotinohidrazida
(21a), (E)-N'-(3-(4-fluoro-fenil)akriloil)isonikotinohidrazida (21b), (E)-N'-(3-(2,5
diklorofenil)akriloil)-isonikotinohidrazida (21c), dan (E)-N'-(3-(2,6-difluorofenil)
akriloil)isonikotinohidrazida (21d) dengan randemen 11-81% tanpa melibatkan
tionil klorida. Senyawa (21a-d) mampu menghambat α-glukosidase yang lebih baik
dibandingkan akarbose (10).

Diabetes mellitus (DM) is a non-communicable disease that is found in type
1 and type 2. Type 2 diabetes is the most common (90-95%) caused by insulin
resistance and impaired insulin secretion. Treatment of type 2 DM is carried out by
controlling blood sugar levels in the form of regulating diet, limiting the amount of
carbohydrate consumption, exercising regularly, and taking drugs as inhibitors of
the α-glucosidase enzyme. The side effects of this use are diarrhea, abdominal pain,
flatulence, hypoglycemia, liver damage, drug resistance, weight gain, heart
problems, and high costs. The amides of cinnamic acid show potential as lead
compounds in the development of -glucosidase inhibitors. The amide synthesis was
carried out by involving thionyl chloride which is included in the list of Chemical
Weapons Conventions, and List-3 Chemicals in the Law of the Republic of
Indonesia Number 9 of 2008 concerning the use of chemicals and the prohibition
of the use of chemicals as chemical weapons; As a result, the research carried out
succeeded in developing a method that did not involve thionyl chloride. Applied in
the synthesis of compounds (E)-N'-(3-phenyl)acryloyl)isonicotinohydrazide (21a),
(E)-N'-(3-(4-fluorophenyl)acryloyl)isocotinohydrazide
(21b),
dichlorophenyl)acryloyl)isonicotinohydrazide
(21c),
(E)-N'-(3-(2,5
and
(E)-N'-(3-(2,6
difluorophenyl)acryloyl)isonicotinohydrazide (21d) were successful, respectively,
synthesized with a yield of 11-80%. The bioactivity test found that the compound
(21a-d) was able to inhibit -glucosidase with an IC50 value of 50-79 µM better than
acarbose (10).
