Abstract

Genangan dapat menyebabkan hasil produksi suatu
tanaman menurun, utamanya pada tanaman terestrial seperti
tembakau. Sehingga, dapat meningkatkan kerugian petani
tembakau. Penelitian ini bertujuan untuk mengetahui respon
Nicotiana tabacum yang dicekam genangan periodik ditinjau dari
aspek anatomi, fisiologi, morfologi dan biokimia. Empat varietas
tanaman tembakau (Jepon Pelakean, Jinten, Manilo dan
Marakot) dicekam genangan periodik pada fase vegetatif dengan
tahapan waterlogging selama lima hari dan dilanjutkan dengan
partial submergence selama lima hari berikutnya. Data
pengamatan dianalisis menggunakan ANNOVA One-Way dengan
taraf kepercayaan 95% untuk menguji parameter morfologi dan
fisiologi. Pada aspek morfologi, Jepon Pelakean memiliki respon
lebih baik pada parameter tinggi tanaman, panjang akar,
diameter batang, lebar daun dan jumlah akar adventif dibanding
ketiga varietas uji lain. Rerata penutupan stomata tertinggi pada
Manilo (95,2%), penurunan klorofil tertinggi terjadi pada Manilo
sebesar (41,5%), dan berat kering tertinggi oleh Jepon Pelakean
sebesar (1,17 mg). Hasil analisis SDS-PAGE pada Jepon
Pelakean, Manilo dan Marakot menunjukkan bahwa terdapat
peningkatan ekspresi protein dengan berat molekul 134,98 kDa
secara konsisten di kondisi waterlogging. Sedangkan, Jinten
menunjukkan ekspresi tersebut saat partial submergence.

Flooding could decrease the yield of any terrestrial plant,
including tobacco. Thus condition may lead to economic losses
for the farmers. The aims of the research are to evaluate the
responses of tobacco (Nicotiana tabacum) plant to periodic
flooding stress based on anatomy, physiology, morphology and
biochemistry aspects. Four varieties of tobacco (Jepon Pelakean,
Jinten, Manilo and Marakot) were treated by flooding stress
during vegetative phase; by means of waterlogging stress for five
days continued by partial submergence for the next five days.
Data were analyzed using One-Way ANNOVA at 95%
confidence level to test the morphological and physiological
aspects. In the morphology aspect, Jepon Pelakean have better
responses on parameters of plant height, root length, stem
diameter, leaf width and number of adventitious roots than the
other three varieties. Furthermore, highest mean of stomatal
closure occurred on Manilo (95.2%), highest chlorophyll
decrement occurred in Manilo (41.5%), and highest dry weight
occurred in Jepon Pelakean (1.17 mg). The results of SDS-PAGE
analysis of Jepon Pelakean, Manilo and Marakot show increased
expression of a protein with molecular weight of 134.98 kDa
consistently in waterlogging treatment. Meanwhile, Jinten
showed that expression in partial submergence.
