Abstract
Calophyllum inophyllum atau yang biasa disebut nyamplung
memiliki banyak khasiat yang dapat dimanfaatkan mulai dari akar, batang, daun, hingga biji. Menurut Ling tahun 2009, daun tanaman ini berfungsi mengobati berbagai macam penyakit seperti iritasi pada mata, penyakit kulit, pembengkakan kaki, disentri, migrain, vertigo,
dan lain-lain. Menurut Thengane et al, 2006, dalam daun nyamplung, di sinyalir mengandung agents chemopreventive cancer, yaitu coumarin dan xanthone sebagai antimicrobial activity. Penelitian ini bertujuan untuk mengetahui senyawa bioaktif yang terkandung pada fraksi polar dan non polar daun nyamplung, cara untuk mengisolasi senyawa xanthone dan coumarin, serta mengetahui kondisi proses pemisahan xanthone dan coumarin yang optimum. Bubuk daun nyamplung diekstraksi dengan pelarut metanol untuk mendapatkan
crude ekstrak daun. Crude ekstrak yang didapatkan kemudian
dipisahkan fraksi polar dan non polarnya dengan ekstraksi liquid-liquid dengan pelarut metanol - petroleum eter. Variabel pelarut total yang digunakan yaitu 10, 30, 50, serta 70 (g/g) dan rasio perbandingan pelarut polar (metanol) dan non polar (petroleum eter) yaitu 25:75 dan
50:50 (g/g). Kemudian dilakukan pula pemisahan secara berkelanjutan (multiple extraction) sampai fraksi polar dan non polar didapatkan. Fraksi polar dan non polar ekstrak daun yang didapatkan kemudian diuji secara kualitatif menggunakan TLC (Thin Layer Chromatography) dan GC-MS (Gas Chromatography-Mass Spectrometry), serta secara kuantitatif menggunakan Spectrophotometry UV-Vis untuk menganalisa kadar coumarin dan xanthone dalam ekstrak daun nyamplung. ====================================================================================== Callophylum inophyllum or also known as nyamplung has many savour which can be used from root, bark, leaves, and seeds. According to Ling, 2009, this nyamplung leaves function is able to cure various
diseases such as eye irritation, skin disease, vertigo, etc. Accoording to Thengane et al, 2006, nyamplung leaves are expected to contain agents chemopreventive cancer, which are coumarin and xanthone as antimicrobial activity. This study aims to identify bioactive compound which is contained in polar and non polar fractions of nyamplung leaves and to determine method to isolate xanthone and coumarin, also to determine the optimum condition for isolating processs of xanthone and coumarin. Nyamplung leaves powder was extracted using methanol to obtain crude extract of nyamplung leaves. Crude extract then was
separated by polar and non polar fraction using liquid-liquid extraction with methanol - petroleum eter solvents. The liquid-liquid extraction was applied with variable of total solvent which was used 10, 30, 50 and 70 (g/g) and the polar (methanol) – non polar (petroleum eter) solvent
ratio 25:75 and 50:50, (g/g). Then obtained too continuing separation multiple extraction) until polar and non polar fraction was available. Polar and non polar fraction of extract leaves that were obtained then tested using TLC (Thin Layer Chromatography) and GC-MS (Gas Chromatography Mass Spectrometry) for qualitative analysis, also
using and Spectrophotometry UV-Vis for quantitative analysis to analyze coumarin and xanthone content in nyamplung leaves extract.
