Abstract

Lipase (EC3.1.1.3) atau disebut juga glycerol ester hydrolases adalah kelas enzim hidrolisis yang memecah trigliserida menjadi asam lemak bebas dan gliserol. Penggunaan karier biopolimer berbasis bahan alam merupakan salah satu alternatif material karena memiliki kelebihan murah, mudah diperoleh dan praktis. Tujuan dari penelitian ini adalah memanfaatkan bahan polimer alami yaitu pati tapioka, gelatin, sodium alginat dan kitosan sebagai karier imobilisasi lipase dengan parameter uji yaitu aktivitas enzim, stabilitas enzim, masa simpan enzim dan analisis gugus fungsi polimer menggunakan FTIR. Hasil penelitian didapatkan bahwa bahan polimer alami dapat dimanfaatkan dalam imobilisasi lipase, dengan hasil lipase terimobilisasi menggunakan biopolimer pati tapioka teraminasi memiliki aktivitas tertinggi sebesar 1.308,7 U/ml dengan kandungan protein 0,207 mg/ml. Hasil analisis FTIR menunjukkan adanya ikatan pada gugus (N-H), (C-N), (C-H) pada lipase terimobilisasi. Pada hasil uji penggunaan kembali dan masa simpan lipase terimobilisasi menggunakan biopolimer pati tapioka teraminasi menunjukkan aktivitas relatif tertinggi yaitu sebesar 69,4% dan 83,7%. ============================================================================== Lipase (EC3.1.1.3) or also known as glycerol ester hydrolases is a class of enzyme to break down the hydrolysis of triglycerides into diglycerides, monoglycerides, free fatty acids and glycerol. The usage of biopolymer-based carrier in enzyme immobilization has advantages in terms of cheap, being obtained easily and practical. The purpose of this study is using natural polymer such as tapioca starch, gelatine, sodium alginate, and chitosan as lipase immobilization carrier which further analysed in enzymatic activity assay, enzyme stability, enzyme storage and polymer functional group analysis using FTIR. Natural polymer could be used for lipase immobilization as a result of this study. Immobilized lipase with aminated tapioca starch biopolymer has the highest activity among all biopolymer which is 1.308,7 U/ml with protein content of 0,207 mg/ml. FTIR result showed a bond formation on (N-H), (C-N), (C-H) group in immobilized lipase. Enzyme recovery and immobilized lipase storage testing with aminated tapioca starch resulted highest relative activity on 96,4% and 83,7%.
