Abstract
Air limbah pabrik tahu mengandung senyawa organik,
yaitu protein, jika kandungan protein air limbah pabrik tahu yang
tinggi dapat menyebabkan kerusakan lingkungan air. Untuk
mencegah terjadinya kerusakan dapat dilakukan dengan
pengurangan kandungan protein air limbah pabrik tahu dengan
memanfaatkan enzim bromelin amobil dengan matriks kitosan
dari cangkang udang. Pada penelitian ini, enzim bromelin
diisolasi dari buah nanas dengan pengendapan menggunakan
larutan ammonium sulfat jenuh dan kasein sebagai substratnya.
Pada uji aktivitas katalitik diperoleh fraksi enzim yang terbesar
pada konsentrasi 30% ammonium sulfat jenuh. Penentuan
kandungan protein menggunakan metode kolorimetri dengan
reagen biuret pada λmaks 520 nm, penelitian ini dilakukan dengan
optimasi jumlah enzim bromelin amobil dan waktu inkubasi.
Hasil dari penelitian menunjukkan bahwa enzim bromelin amobil
dengan kitosan mampu mendegradasi protein dalam air limbah
pabrik tahu optimum pada penggunaan 2 mg enzim bromelin
amobil selama 10 jam inkubasi dapat mendegradasi protein
89,506% (7,91377 mg).

Liquid waste of tahu industry contains organic
compounds, that is protein, if protein level of liquid waste of tahu
industry was so high, its can cause some trouble in environment
of water. To prevent damage, its could be done with the reduction
of protein to liquid waste of tahu industry by using immobilize of
bromelain enzyme using matrix of chitosan from shrimp shells. In
this research, bromelain enzyme was isolated from pineapple by
precipitation of ammonium sulphate and casein as substrate. The
test of catalytic activity was got the biggest enzyme fraction of
30% ammonium sulphate concentrate. Determination of organic
compounds was used colorimety method by biuret at λmaks 520
nm, in this research was conducted by optimization of the amount
of immobilize bromelain enzyme and incubation time. The
research showed that immobilize bromelain enzyme using matrix
of chitosan can degrade the biggest protein compounds of liquid
waste of tahu industry using 2 mg immobilize bromelain enzyme
for 10 hours incubation, was 89.506% (7.91377 mg).
