Abstract
Pencemaran tanah oleh hidrokarbon dapat berasal dari kegiatan eksplorasi dan eksploitasi minyak bumi yaitu banyaknya tumpahan atau ceceran dari proses pengeboran
dan kebocoran pipa. Produk turunan minyak bumi yang paling banyak digunakan adalah bahan bakar solar. Solar merupakan salah satu produk utama dari penyulingan minyak bumi yang menjadi sumber utama polusi di lingkungan karena merupakan hidrokarbon kompleks. Kelarutan hidrokarbon yang rendah dalam tanah menyebabkan rendahnya efektifitas biodegradasi. Salah satu cara menanggulangi permasalahan ini adalah dengan bioremediasi. Terdapat dua
pendekatan yang dapat digunakan dalam bioremediasi tumpahan minyak, yaitu biostimulasi dan bioaugmentasi. Penelitian ini bertujuan untuk mengkaji pengaruh metode biostimulasi yang dikombinasikan dengan metode bioaugmentasi dalam mendegradasi senyawa hidrokarbon minyak solar dalam tanah.
Pada penelitian ini variabel yang digunakan adalah jenis kultur campuran bakteri (Bacillus subtilis -
Pseudomonas fluorescens; B. subtilis - Pseudomonas
putida; P.putida–P. fluorescens), variasi konsentrasi penambahan inokulum 5% (b/b) dan 10 % (b/b), dan variasi konsentrasi penambahan nutrien anorganik berupa pupuk urea dan superfosfat (100:10:10 dan 100:5:1). Penelitian ini dilakukan dalam skala laboratorium dengan konsentrasi penambahan minyak solar 10% (b/b). Uji utama adalah uji bioremediasi tanah tercemar hidrokarbon dengan pengukuran parameter yang diuji meliputi kadar minyak yang terlarut (Total Petroleum Hydrocarbon), suhu, pH, total koloni mikroba serta pengukuran komponen hidrokarbon dengan Gas Chromatography-Mass Spectrometry.
Hasil penelitian menunjukkan penurunan nilai TPH terbaik pada campuran P. fluorescens - P. putida dengan konsentrasi inokulum 10% b/b yaitu 58.131 mg/kg (H
-15) dengan tingkat degradasi sebesar 62,35%. Sedangkan berdasarkan variasi rasio pupuk anorganik,
P. fluorescens - P. putida menunjukkan hasil
penurunan nilai TPH tertinggi pada rasio penambahan C:N:P 100:5:1 penurunan TPH sebesar 54.968 mg/kg (H
-15) dengan tingkat degradasi sebesar 64,04%. ==================================================================================================
Pollution of soil by hydrocarbon can be caused by exploration and exploitation activities of petroleum that is the number of spills or spills from the drilling process and leakage of pipes. The most widely used petroleum derivative product is diesel fuel. Solar is one of the main products of petroleum refining which is the main source of pollution in the environment because it is a complex hydrocarbon. The low hydrocarbon solubility in the soil causes the low effectiveness of biodegradation. One way to overcome this problem is by bioremediation. There are two approaches that can be used in bioremediation of oil spills, namely biostimulation and bioaugmentation. This study aims to examine the effect of biostimulation method combined with bioaugmentation method in degrading hydrocarbon compounds in the soil.
In this study the variables used were mixed culture bacteria (Bacilus subtilis-Pseudomonas fluorescens, B. subtilis - Pseudomonas putida, P. putida – P. fluorescens), variation of inoculum addition concentration were 5% and 10% (w/w), and variations in the concentration of inorganic fertilizers addition in the form of urea and superphosphate (100: 10: 10 and 100: 5: 1). This research was conducted on a laboratory scale with diesel concentration added 10% (w/w). The main test was hydrocarbon contaminated soil bioremediation test with measurement of tested parameters including Total Petroleum Hydrocarbon, temperature, pH, total microbial colonies and measurement of hydrocarbon components with Gas Chromatography-Mass Spectrometry.
The results showed that the best removal of TPH value on the mixture of P. fluorescens - P. putida in inoculum 10% (w/w) was 58.131 mg/kg (H-15) with the degradation rate of 62.35%. Meanwhile, based on variation of ratio of inorganic fertilizer, P. fluorescens - P. putida showed the highest decrease of TPH value on the addition ratio of C: N: P 100: 5: 1 decrease of TPH by 54.968 mg/kg (H-15) with degradation level equal to 64,04 %.
