Abstract

Mikroalga B.braunii adalah bahan baku penghasil lipid yang cukup potensial. Mutasi dengan menggunakan sinar UV-B merupakan metode untuk meningkatkan produktivitas mikroalga. Pengurangan kadar nitrogen dalam nutrien dan lama waktu kultur dilakukan untuk mengetahui pengaruhnya terhadap produktivitas lipid dan lipid content mikroalga B.braunii.
Variabel nitrogen dalam substrat dibuat dengan komposisi NaNO3 100 mg/L dan NaNO3 0,03 mg/L. Prekultur dilakukan selama 6 hari dengan pemberian nutrien normal setiap 24 jam. Tahap selanjutnya adalah kultur mikroalga yang dilakukan selama 7 hari dan 20 hari dengan pemberian nutrien sesuai variabel. Kondisi operasi meliputi penambahan aerasi sebanyak 2,5 L/menit, CO2 sebanyak 50 mL/menit dan siklus pencahayaan (terang : gelap) 24 jam : 0 jam. Pertumbuhan mikroalga dihitung dengan menggunakan metode counting chamber setiap 24 jam. Untuk mendapatkan lipid digunakan ekstraksi menggunakan n-heksan.
Berdasarkan penelitian yang dilakukan didapatkan hasil bahwa kadar nitrogen dalam nutrien sangat berpengaruh terhadap perrtumbuhan dan kadar lipid mikrolaga B.braunii. Mikroalga B.braunii alami dengan nutrien normal memberikan pertumbuhan sel paling pesat dibanding mikroalga B.braunii termutasi UV-B. Produktivitas biomassa dan lipid sangat ditentukan oleh pertumbuhan jumlah sel dan massa sel mikroalga B.braunii. Produktivitas biomassa tertinggi dihasilkan B.braunii alami dengan normal nutrien waktu kultur 7 hari yaitu sebesar 0,235 (mg/mL)/ hari. Produktivitas lipid paling tinggi dihasilkan oleh B.braunii mutasi UV-B rendah nitrogen waktu kultur 7 hari yaitu sebesar 0,09 (mg/mL)/ hari. Lipid content paling besar yaitu B.braunii alami dan mutasi UV-B rendah nitrogen waktu kultur 20 hari sebesar 67,42% dan 70,03%. B.braunii alami memberikan produktivitas biomassa tertinggi. Produktivitas lipid tertinggi dihasilkan oleh B.braunii mutasi UV-B, hal ini menunjukkan bahwa perlakuan mutasi UV-B mampu meningkatkan produktivitas lipid. Hasil analisa GC menunjukkan kandungan asam lemak bebas (FFA) dan Monoacyglycerol (MAG) lebih besar dibandingkan komponen lainnya. Kandungan FFA dan MAG untuk variable rendah nitrogen yang terbesar adalah B.braunii mutasi dengan waktu kultur 7 hari yaitu sebesar 22,54 % dan 38,289 sementara kandungan Triacyglycerol (TAG) sebesar 1,006%.

B.braunii is a microalgae that currently being tested for it’s ability because of high lipid content.UV-B radiation is a method to increase productivity of microalgae. Reduced levels of nitrogen in nutrients and duration of culture were conducted to determine the effect on lipid productivity and lipid content of B. braunii.
The nitrogen variables in substrate were prepared with the composition of NaNO3 100 mg/L and NaNO3 0,03 mg/L. Pre-cultivation microalgae B.braunii in 6 days with normal nutrient every 24 hour. Cultivation time done in 7 days and 20 days with nitrogen in NaNO3 varied, aerationaddition 2,5 L/min, CO2 addition 50 mL/min and a light cycle ratio (light: dark) is 24 hours : 0 hours. Microalgae growth is calculated with counting chamber every 24 hours. Extraction of lipid using n-hexane to obtain the lipid in B.braunii.
It was found that the effect of nitrogen content in nutrients influenced the growth and lipid content of B.braunii. The natural B. braunii with normal nutrients provides the most rapid cell growth compared to UV-B mutation. The productivity of biomass and lipids is determined by the growth of cells and the mass of cells B.braunii. The highest biomass productivity was produced by natural B.braunii with normal nutrient at 7 days cultivation time which 0,235 (mg / mL) /day. The highest productivity of lipids produced by B.braunii UV-B mutation low nitrogen at 7 days cultivation which 0,09 (mg / mL) /day. The largest lipid content was natural B.braunii and UV-B mutation with low nutrient at 20 days cultivation time were 67,42% and 70,03%. Natural B.braunii provide the highest biomass productivity. The highest lipid productivity was generated by B.braunii UV-B mutations. Indicating of this result, UV-B mutation can increase the productivity of lipids. The result of GC analysis showed that free fatty acid content (FFA) and Monoacyglycerol (MAG) were bigger than other components. The content of FFA and MAG for the largest variable of low nitrogen is B.braunii UV-B mutation with 7 days cultivation time which 22,54% and 38,289 while Triacyglycerol content (TAG) equal to 1,006%.
