Abstract

Motivasi utama penelitian ini adalah merancang suatu sistem multispectral imaging (MSI) untuk membantu dermatologis dalam memperoleh informasi berupa kandungan melanin dan darah pada kulit. Beberapa studi sebelumnya menunjukkan bahwa MSI cocok untuk digunakan dalam kajian dermatologi karena sistem ini mampu memperoleh informasi spektral dan spasial. Penulis telah berhasil merancang, melakukan karakterisasi, dan memvalidasi suatu sistem MSI yang nantinya dapat digunakan pada kajian dermatologi dengan pendekatan baru. Sistem yang dirancang terdiri dari dua bagian utama: sumber iluminasi dengan LED untuk 12 panjang gelombang tertentu dan sistem akuisisi gambar yang menggunakan kamera monokromatik yang memiliki resolusi 640x480 piksel dengan lensa makro objektif dengan perbesaran 80-130 kali. Penulis juga menggunakan Artificial Neural Networks (ANN) untuk merekontruksi kubus reflektansi sehingga sistem MSI mampu menghadirkan data reflektansi pada panjang gelombang 380-880 nm dengan kenaikan langkah sebesar 5 nm. Untuk estimasi kandungan melanin dan kandungan darah, teori propagasi cahaya Kubelka-Munk pada dua layer digunakan untuk memodelkan reflektansi total dari kulit manusia. Dengan kata lain, reflektansi total dimodelkan sebagai fungsi kandungan melanin dan darah. Dengan melakukan fitting atau meminimalkan nilai error antara data reflektansi yang terukur oleh sistem MSI dan data reflektansi dari simulasi model, maka kandungan melanin dan darah bisa diestimasi secara akurat. Penulis telah melakukan validasi dengan mengakuisisi data reflektansi dengan MSI dan membandingkannya dengan data dari spektrometer. Hasil estimasi kandungan melanin dengan menggunakan sistem MSI dan spektrometer mengikuti persamaan y = 0,9565x dengan koefisien determinasi (R2) sebesar 0,9934. Di sisi lain, hasil estimasi kandungan darah mengikuti persamaan y = 1,1881x dengan koefisien determinasi (R2) sebesar 0,9226. Sistem MSI juga telah berhasil diuji secara in vivo untuk mengakuisisi data reflektansi kulit tangan manusia ukuran ± 10 mm x 7,5 mm dengan resolusi sebesar 48 piksel/mm2. Rata-rata hasil estimasi kandungan melanin dan darah secara berturut memiliki perbedaan sebesar 1,29% dan 1,06% v/v dibandingkan dengan nilai yang diestimasi dengan Spektrometer USB650. Dengan temuan ini, sistem MSI yang telah penulis rancang bisa lebih lanjut digunakan untuk assesment kandungan melanin dan darah pada kulit secara langsung dan non-invasif.
==================================================================================================================The main motivation of this research is to design a multispectral imaging (MSI) system to assist dermatologists in obtaining information in the form of melanin content and blood on the skin. Several previous studies have shown that MSI is suitable for use in dermatology studies because the system is capable of obtaining spectral and spatial information. We have successfully designed, characterized, and validated an MSI system that could later be used in dermatology studies with new approaches. The designed MSI system consists of two main parts: an illuminated source with LEDs for 12 specific wavelengths and an image acquisition system using a monochromatic camera that has a resolution of 640x480 pixels with an objective macro lens with magnification of 80-130 times. We also use Artificial Neural Networks (ANN) to reconstruct the reflectance cube so that the MSI system is able to present reflectance data at 380-880 nm wavelengths with a step increase of 5 nm. To estimate the content of melanin and blood content, Kubelka-Munk light propagation theory on two layers is used to model the total reflectance of human skin. In other words, the total reflectance is modeled as a function of melanin and blood content. By fitting the curve or minimizing the error value between the reflectance data measured by the MSI system and the reflectance data from the model simulation, the content of melanin and blood can be accurately estimated. We have validated the system by acquiring the reflectance data with MSI and comparing it with the data from the spectrometer. Melanin content estimation result using MSI system and spectrometer follow the equation y = 0.9565x with the coefficient of determination (R2) equal to 0.9934. On the other hand, the result of the blood content estimation follows the equation y = 1.1881x with the coefficient of determination (R2) of 0.9226. The MSI system has also been successfully tested in vivo to acquire the hand-skin reflectance data of ± 10 mm x 7.5 mm in size with a resolution of 48 pixels / mm2. The mean estimate of melanin and blood content was 1.29% and 1.06%, respectively, compared to the values estimated with the Spectrometer USB650. With these findings, the MSI system we have designed could be further used for the direct and non-invasive assessment of melanin and blood content of the skin.
