Abstract

Selama ini pemuliaan budidaya tembakau varietas somporis masih dilakukan secara konvensional. Pemuliaan secara konvensional sering mengalami kendala seperti masa produksi yang lebih lama dan hasil yang diperoleh tidak seragam. Oleh karenanya diperlukan alternatif teknik pemuliaan budidaya tembakau yang lebih cepat dan efisien. Salah satu teknik yang dapat dikembangkan adalah induksi ginogenesis untuk membentuk tanaman haploid menggunakan pollen yang diberi penyinaran sinar gamma. Tahap preparasi pollen sebelum dilakukan penyinaran sinar gamma sangat penting dilakukan. Proses pembibitan tanaman tembakau mulai dari benih, fase vegetatif hingga fase generatif dilakukan sesuai dengan panduan budidaya tanaman tembakau. Bunga yang digunakan untuk pemanenan pollen adalah bunga yang memiliki ciri panjang bunga ± 5 - 5,5 cm, ujung mahkota bunga sedikit membuka dan berwarna hijau kekuningan. Pollen yang didapat disimpan pada suhu 4 oC selama 3 hari untuk menunggu tanaman donor berbunga. Hasil penelitian menunjukan tanaman tembakau varietas somporis menghasilkan rata-rata 5,5 mg pollen per bunga. Setelah dipolinasi pada tanaman donor, pollen yang telah disimpan selama 3 hari pada suhu 4 oC memiliki viabilitas yang tinggi. Hal ini dapat dilihat dari persentase pembentukan polongnya yang mencapai 100%.============ During this time, breeding cultivation of somporis varieties tobacco plant is still done conventionally. Conventional breeding often faces some obstacles such as longer production period and various result. In order to solve the obstacles, an efficient and faster alternative technique of tobacco breeding was needed. One of the technique is gynogenetic induction to produce haploid plant using gamma ray irradiated pollen. The preparation stage of the pollen before gamma ray irradiation was essential. Tobacco seedling process began with seed, vegetative phase until generative phase was done according to the guidelines of tobacco cultivation. The flowers were used for pollen harvesting had some characteristics such as ± 5 - 5,5 cm of long, the crown tip of the flower was slightly open, and its color was yellowish green. The pollen was stored at 4 oC for 3 days to wait for flowering donor plants. the result showed that somporis varieties tobacco plant produced an average of 5,5 mg pollen for each flower. After being pollinated on donor plant, the pollen that had been stored for 3 days at 4 oC had high viability. It can be seen from the seeds set percentage of donor plant that pollinated with preparated pollen which reached 100 %.
