Abstract

Kanker merupakan penyakit yang menyebabkan kematian
terbesar di dunia. Sejauh ini pengobatan kanker dapat dilakukan dengan melakukan pembedahan, radioterapi, kemoterapi dan terapi hormon. Namun, efek samping dari obat kemoterapi tidak hanya menghancurkan sel kanker tetapi juga menyerang sel-sel normal. Penggunaan bahan alam mampu meminimalkan efek samping dari obat sintesis. Senyawa Trisindolina 1 merupakan bahan alam yang telah diuji sitotoksisitasnya terhadap sel kanker dan menunjukkan hasil yang positif tetapi belum diketahui bagaimana aktivitasnya terhadap induksi aktifitas fagositosis.
Pada penelitian ini dilakukan uji In vitro senyawa Trisindolina 1 terhadap aktivitas fagositosis sel makrofag Mencit (Mus musculus). Makrofag didapatkan dari peritoneal Mencit yang dikulturkan pada medium RPMI (Roswell Park Memorial Institute). Hasil kultur ditambahkan dengan Latex Beads sebagai model antigen dalam proses fagositosis sel makrofag, kemudian ditambahkan senyawa Trisndolina 1 dengan konsentrasi 100 µg/mL, 50 µg/mL, 25 µg/mL, 12,5 µg/mL, 6,25 µg/mL. Hasil dari penelitian ini menunjukkan bahwa pada konsentrasi 6,25 µg/mL senyawa trisindolina 1 mampu menginduksi kapasitas dan indeks fagositosis sel makrofag yang paling optimal yaitu sebesar 38,34 dan 2,74.
============= Cancer is a disease causes the highest mortality rate. Current cancer treatment can be done by surgery, radiotherapy, chemotherapy and hormone therapy. However, chemotheraphy agent can shows some side effects such as not only the damage of cancer cells but also the damage of normal cells. The use of natural compound is capable of producing effects of synthetic drugs. Trisindolina 1 compounds become a natural substance that has proven its cytotoxicity to cancer cells and show positive results but the ability of this compound to induce phagocytosis of macrophage still remain unknown.
In this research, Trisindolina 1 is used on macrophage of Mus musculus to measure phagocytosis activity in vitro. Macrophages are obtained from Peritonel Mice cultured on RPMI (Roswell Park Memorial Institute) medium. Cultures were added with Latex Beads as antigent modeling and Trisndolina 1 were added at concentrations of 100 μg/mL, 50 μg/mL, 25 μg/mL, 12.5 μg/mL, 6.25 μg/mL, respectively. The results of this study indicate that the trisindoline 1 compound is able to induce the optimal activity of phagocytosis on macrophage with concentration 6.25 μg/mL (capacity = 38,34, and index = 2,74).
