Abstract
Kurkuminoid yang terkandung di dalam kunyit (Curcuma
domestica valeton) merupakan bahan aktif yang dapat berperan
sebagai antibakteri. Kurkuminoid mempunyai aktivitas
menghambat proses proliferasi sel bakteri dan diduga sebagai
target penting dari antibiotik. Kurkuminoid yang merupakan
senyawa polyphenolic ini mampu mempengaruhi fungsi membran
sel bakteri yang berperan sebagai barier terhadap enzim otolitik,
dengan memodifikasi lipid bilayer membran sel bakteri.
Kurkuminoid dan protocatechuic acid yang merupakan senyawa
polyphenolic berkerjasama dengan mekanisme kerja antimikroba,
yaitu melalui penghambatan fungsi membran sel bakteri dan
penghambatan sintesis protein pada sel bakteri. Senyawa
polyphenolic ini memiliki aktivitas antibakteri dan memiliki daya
hambat yang baik dalam konsentrasi yang kecil. Tahap uji
antimikroba diuji menggunakan kurkuminoid yang dilarutkan
dalam tiga jenis NADES yang berbeda yaitu: MAS-H2O (1:1:18);
FG-H2O (1:1:7); dan FS-H2O (2:1:15) pada dua jenis mikroba: E.
coli (gram negatif) dan S. aureus (gram positif). Uji Minimum
Inhibitory Concentration (MIC) dan Minimum Bactericidal
Concentration (MBC) dilakukan dengan konsentrasi kurkuminoid
bervariasi ( 0 mM; 2 mM; 2,25 mM; 2,5 mM; 2,75 mM; 3 mM;
3,25 mM; 3,5 mM; 3,75 mM; dan 4 mM) dan konsentrasi suspensi
bakteri uji dengan kepadatan 108 sel/ml sebanyak 1 mL dengan
waktu inkubasi 1x24 jam dan suhu inkubasi 37 oC. Uji antimikroba
juga dilakukan terhadap masing-masing komponen murni penyusun NADES: sukrosa, malic acid, fruktosa, dan glukosa.
NADES MAS-H2O memiliki aktivitas anti-mikroba paling efektif
dibandingkan dengan tipe netral (FG-H2O dan FS-H2O). Toksisitas
NADES MAS-H2O terhadap E. coli dan S. aureus dipengaruhi oleh
pH larutan NADES, dalam hal ini adalah malic acid. Asam malat
adalah asam kuat (pH < 3), sementara NADES mengandung gula
memiliki pH lebih tinggi (pH > 5) dan tidak memberikan aktivitas
baik MIC maupun MBC. ========================================================================================================
Curcuminoids, a bioactive compound from turmeric (Curcuma
domestica valeton) has antibacterial effect. It is also indicated has
an inhibitory effect to the bacterial cell proliferation; hence
potentially use as an important target of antibiotics. Curcuminoids,
a polyphenolic compound able to affect the function of bacterial
cell membranes that act as a barrier to the autolithic enzymes, by
modifying the lipid bilayer of bacterial cell membranes.
Curcuminoids and protocatechuic acid which is also a
polyphenolic compound in cooperation with antimicrobial action
mechanisms, namely through inhibition of bacterial cell membrane
function and inhibition of protein synthesis in bacterial cells. These
polyphenolic compounds have antibacterial activity and have good
drag in small concentrations. The antimicrobial test stage was
conducted using curcuminoids dissolved in three different types of
NADES: MAS-H2O (1:1:18); FG-H2O (1:1:1); and FS-H2O
(2:1:15) in two types of microbes: Escherichia coli (gramnegative)
and Staphylococcus aureus (gram-positive). Minimum
Inhibitory Concentration and Minimum Bactericidal
Concentration were tested with concentrations of curcuminoids (0
mM; 2 mM; 2.25 mM; 2.5 mM; 2.75 mM; 3 mM; 3.25 mM; 3,5
mM; 3.75 mM; and 4 mM), concentration of bacterial suspension
about 108 cells/ml of 1 mL with incubation time of 1x24 hours and
incubation temperature 37oC. Antimicrobial tests were also
conducted on each of the pure composite components of NADES, such as: sucrose; malic acid; fructose; and glucose. NADES (MASH2O)
was the most effective anti-microbial activity compared to
pure types (FG-H2O and FS-H2O). The toxicity of NADES to a
microorganism might be affected by the pH of NADES itself.
NADES contained malic acid was a strong acid (pH <3), while
NADES contained sugar was weak acid (pH >5). The active
compound of curcuminoids had broad spectrum of antibacterial
activity was active antibacterial against various types of gram
positive and gram negative bacteria.
