Abstract

Metilen Biru (MB) merupakan salah satu pewarna yang umum digunakan dalam industri. Penghilangan zat warna MB yang efektif dapat berlangsung secara biologis menggunakan aktivitas mikroorganisme. Pada penelitian ini metilen biru dihilangkan melalui proses biodekolorisasi oleh kultur campuran jamur pelapuk putih Phlebia brevispora dan bakteri Pseudomonas aeruginosa. Proses biodekolorasi MB dengan konsentrasi 100 mg/L diamati selama 7 hari di dalam media cair Potato Dextrose Broth (PDB) dengan variasi penambahan volume P. aeruginosa sebanyak 2, 4, 6, 8, dan 10 mL terhadap 1 mL P. brevispora. Analisis dekolorisasi metilen biru diukur menggunakan instrumen UV-Vis dan metabolit produk yang dihasilkan organisme dianalisis menggunakan instrumen LC-MS/TOF. Kultur campuran mampu mendekolorisasi MB sebesar 39,28% sedangkan dekolorisasi menggunakan P. brevispora mencapai 1,58%. Metabolit yang dihasilkan dari proses dekolorisasi MB adalah C16H18N3O5S.======================================================================================================Dye is compound with high stability which caused by its complex aromatic structure. It makes dye to potentially become a water pollutant and causes negative effects on organism. In this study, MB was removed by biodecolorization process using white-rot fungus Phlebia brevispora and Pseudomonas aeruginosa mixed culture. P. aeruginosa with volume 2, 4, 6, 8, 10 mL and 100 mg / L MB were added in 1 mL P. brevispora. The biodecolorization was carried out in Potato Dextrose Broth (PDB) medium and was observed in 7 days. MB decolorization was measured using UV-Vis instrument and product metabolites produced by the organisms were analyzed using LC-MS / TOF instruments. P. aeruginosa and P.brevispora mixed culture able to decolorize MB up to 39,28%, meanwhile using P. brevispora pure culture is only up to 1,58%. Metabolites produced during decolorization MB by mixed cultures are C16H18N3O5S.
