Abstract
Kitosan adalah produk deasetilasi kitin yang mempunyai
banyak manfaat di berbagai bidang. Namun, selama ini solubilisasi kitin
menjadi kitosan dilakukan melalui proses secara kimiawi yang tidak
ramah lingkungan. Permasalahan dalam penelitian ini adalah
bagaimana mengoptimalkan produksi kitin deasetilase sebagai agen
biologi solubilisasi kitin oleh bakteri Bacillus sp. SK II-5 dengan
memanfaatkan limbah perikanan.
Pada penelitian ini dilakukan optimasi produksi kitin
deasetilase pada suhu, pH dan medium produksi enzim. Kandungan
protein diukur berdasarkan metode Bradford dengan BSA (Bovine
Serum Albumin) sebagai standar. Serta dilakukan purifikasi enzim
dengan metode presipitasi ammonium sulfat bertingkat kemudian
protein kitin deasetilase dikarakterisasi dengan pengukuran terhadap
titik isoelektrik, aktivitas enzim, kandungan protein dan elektroforesis
SDS-PAGE.
Hasil penelitian diperoleh aktivitas enzim paling
optimum dihasilkan dari kombinasi medium kulit udang sebagai
sumber karbon, pH 7 dan inkubasi pada suhu 60ºC dengan
aktivitas enzim tertinggi hasil purifikasi amonium sulfat pada
fraksi 60-75% sebesar 969,14 U/ml dengan kandungan protein
0,0024 mg/ml. Kitin deasetilase dari Bacillus sp. SK II-5 memiliki
titik isoelektrik pada pH 5 dan berat molekul 45 kDa

Chitosan is a product of deacetylation chitin which has
many benefits in various fields. Solubilization of chitin into
chitosan has been done through chemical process which is
harmful to the environment. The current study approaches the
problem through green technology in which chitin deacetylase is
optimized by Bacillus sp. SK II-5 as biological agent of chitin
solubilization.
The production optimization of chitin deacetylase
conducted in this study include the temperature, pH and enriched
medium. The protein content was measured by Bradford method
with BSA (Bovine Serum Albumin) as standard. The enzyme was
purified by ammonium sulfate precipitation and characterized by
measuring the isoelectrical point, enzyme activity, protein content
and molecular weight.
Optimum enzyme activity was achieved through
combination of medium shrimp shells as the carbon source with
pH 7 at 60ºC. with the highest enzyme activity results of
purification of ammonium sulfate at 60-75% fraction of 0.00528
U / ml with a protein content of 0.0024 mg / ml. Chitin
deacetylase from Bacillus sp SK II-5 has an isoelectric point at
pH 5 and molecular weight 45 kDa
