Abstract

Tembakau (Nicotiana tabacum) merupakan tanaman komersial bukan pangan yang paling banyak dibudidayakan khususnya di Indonesia. N. tabacum mengandung senyawa aktif dari golongan alkaloid, flavanoid, saponin, dan terpenoid.. Senyawa aktif yang dimiliki pada tembakau dapat digunakan sebagai bioinsektisida yaitu sebagai penangkal serangga salah satunya adalah Plutella xylostella. Tujuan dari penelitian ini adalah mengetahui pengaruh ekstrak daun tembakau terhadap mortalitas P. xylostella serta mengetahui berapa konsentrasi efektif ekstrak daun tembakau (N. tabacum) yang digunakan sebagai bioinsektisida untuk pengendalian hama P. xylostella.
Metode ekstraksi yang digunakan dalam penelitian ini adalah metode maserasi (proses peluruhan senyawa bioaktif pada daun) serta metode aplikasi insektisida pakan menggunakan metode celup. Rancangan penelitian yang digunakan adalah rancangan acak lengkap (RAL) dan parameter uji yang diamati adalah mortalitas P. xylostella. Analisa data dilakukan menggunakan analisis probit, ANOVA One Way serta uji lanjutan DMRT (Duncan Multiple Range Test).
Hasil penelitian menunjukkan bahwa berdasarkan analisis probit LC50 konsentrasi terendah yang efektif terhadap mortalitas larva P. xylostella diatas 50% adalah pada konsentrasi 40%. Serta diikuti konsentrasi yang lebih tinggi 45%, 55% , dan pada konsentrasi 60% memiliki mortalitas 100%. Namun pada konsentrasi 50% menghasilkan mortalitas yang rendah dikarenakan kemampuan larva untuk menghindari senyawa toksik sehingga tidak menyebabkan kematian. Pada penelitian ini tingkat mortalitas berbanding lurus dengan konsentrasi ekstrak karena semakin tinggi konsentrasi maka daya bunuh larva uji juga semakin tinggi.

Tobacco (Nicotiana tabacum) is the most widely cultivated non-food commercial plant also needed especially in Indonesia. N. tabacum contains active compounds from alkaloid groups, flavanoids, saponins, and terpenoids. The active compounds in tobacco can be used as bioinsecticides which are antidotes to one of them is Plutella xylostella.. The purpose of this study was to determine the effect of tobacco leaf extract on mortality of P. xylostella and find out how effective the concentration of tobacco leaf extract (N. tabacum) was used as a bioinsecticides for controlling P. xylostella pests.
The extraction method used in this study was maceration method (decay of bioactive compounds on leaves) and insecticide application method using the dye method. The design used was a complete randomized trial (CRD) and the parameters used were mortality of P. xylostella. Data analysis was performed using probit analysis, ANOVA One Way and DMRT follow-up tests (Duncan Multiple Range Test).
The results showed that based on probit analysis LC50 the lowest concentration that was effective against mortality of P. xylostella larvae was above 50% at a concentration of 40%. With a concentration of 45% higher, 55%, and at a concentration of 60% it has a 100% mortality. However, at a concentration of 50% it produces low mortality due to the ability of larvae to release toxic competencies so as not to cause death. In this study, the mortality rate was directly proportional to the higher concentration of extract so that it increased the larval test also higher.
