Abstract
Sebesar 30% residu lipid dari limbah hasil pengolahan biji Moringa oleifera dapat dimanfaatkan sebagai substrat produksi lipase. Namun, senyawa fenol yang terdapat di dalam limbah sulit didegradasi dan bersifat anti mikroba sehingga perlu dihilangkan. Penelitian ini bertujuan untuk mengetahui agen pretreatment terbaik dalam menghilangkan kandungan fenol pada limbah biji M. oleifera sehingga dapat digunakan sebagai substrat produksi lipase. Beberapa agen pretreatment yang digunakan antara lain lakase, Laccase Mediator System (ABTS), H2SO4, H2O2, dan NaOH. Lipase diproduksi menggunakan isolat Bacillus sp. SK II-5, kemudian dikarakterisasi dengan uji kandungan protein (Bradford), uji titik isoelektrik, dan uji aktvitas secara kualitatif. Hasil dari penelitian ini adalah lakase merupakan agen pretreatment terbaik yang dapat menurunkan konsentrasi total fenol pada limbah M. oleifera sebesar 70% menjadi 3,87 mg GAE/g. Hasil tersebut diikuti dengan tingginya konsentrasi total protein sebesar 0,43 mg/ml serta adanya aktivitas lipase pada rentang 100-200 ppm formaldehid

As much as 30% of lipid residue from Moringa oleifera seeds waste can be used as a substrate for lipase production. However, the phenolic compounds contained in the waste are difficult to degrade and have antimicrobial property which is need to be removed. This study aims to determine the best pretreatment agent in removing phenolic content in M. oleifera seed waste so that it can be used as a substrate for lipase production. Some pretreatment agents used include Laccase, Laccase Mediator System (ABTS), H2SO4, H2O2, and NaOH. Lipase is produced using Bacillus sp. SK II-5 isolates, then characterized by protein content test (Bradford), isoelectric point test, and qualitative activity test. The result of this study is that laccase is the best pretreatment agent that can reduce total phenol concentration in M. oleifera waste by 70% to 3.87 mg GAE / g. The results were followed by a high total protein concentration of 0.43 mg / ml and lipase activity in the range of 100-200 ppm formaldehyde
