Abstract

Perlakuan awal oksidasi laccase pada limbah kulit kopi dalam penelitian ini diharapkan dapat meningkatkan kualitas kompos yang dihasilkan dengan menghilangkan senyawa fenol kulit kopi yang dapat bersifat toksik bagi mikrooganisme. Tujuan dari penelitian ini adalah mengetahui pengaruh perlakuan awal oksidasi laccase pada limbah kulit kopi (Coffea arabica) terhadap kandungan fenol total limbah kulit kopi dan kualitas kompos yang dihasilkan. Optimasi poduksi laccase dilakukan pada 5 jenis medium fermentasi. Uji kadar fenol total, gula pereduksi dan analisis kadar N, P, K, C kompos dilakukan dengan metode spektrofotometri. Hasil yang diperoleh adalah aktivitas laccase tertinggi diperoleh dari medium E yaitu sebesar 1081,2 U/ml. Kadar fenol total dan gula pereduksi terendah diperoleh pada limbah kulit kopi dengan perlakuan awal oksidasi laccase dan ABTS yaitu sebesar 0,007 ± 0,07 mg/ml dan 3,29 ± 0,19 ppm. Perlakuan awal oksidasi laccase tidak berpengaruh terhadap kadar bahan organik. Nilai TPC kompos pada sampel limbah kulit kopi dengan perlakuan awal oksidasi laccase menunjukkan nilai yang lebih stabil dibandingkan kontrol. Namun, perlakuan awal oksidasi ternyata tidak memberikan pengaruh terhadap kadar N, P, K, C, dan rasio C/N kompos yang dihasilkan apabila dibandingkan dengan kontrol.

Pretreatment of laccase oxidation on coffee pulp and husk in this study is expected to improve the quality of compost produced by removing phenol compounds of coffee pulp and husk which can be toxic for microorganisms of composting. The purpose of this study was to determine the effect of pretreatment of laccase oxidation on coffee skin waste (Coffea arabica) on the total phenol content of coffee skin waste and the quality of compost produced. Optimization of laccase production was carried out on 5 types of fermentation medium. Test of total phenol, reducing sugar and analysis of compost N, P, K, C content was conducted by using spectrophotometric methods. The results showed that the highest laccase activity was 1081.2 U / ml which was obtained from the E medium. The lowest total phenol and reducing sugar content were 0.007 ± 0.07 mg / ml and 3.29 ± 0.19 ppm respectively, which were obtained from coffee pulp and husk with laccase oxidation and ABTS pretreatment. Pretreatment of laccase oxidation did not affect the levels of organic matter. The compost TPC value in coffee pulp and husk samples with laccase oxidation pretreatment showed a more stable value than the control. However, the initial oxidation treatment did not have an effect on the levels of N, P, K, C and C/N ratio of compost produced when compared to the controls.
