Abstract

Emas atau Aurum (Au) merupakan salah satu limbah yang dihasilkan oleh industri elektroplating yang dapat bersifat toksik bagi beberapa organisme, namun beberapa bakteri mampu mentolerir. HAuCl4 merupakan senyawa kimia yang mengandung Au. Penelitian ini bertujuan untuk mengetahui ketahanan bakteri Gram positif terhadap HAuCl4 dan perbedaan migrasi proteinnya dengan elektroforesis SDS-PAGE. Penelitian diawali dengan uji resistensi isolat Bacillus cereus S1, Lactobacillus sp. dan Staphylococcus aureus menggunakan medium MSM broth+glukosa 2% mengandung HAuCl4 0,1 ppm; 1 ppm; 5 ppm; dan 10 ppm. Kultur diinkubasi selama 24 jam. Kemudian dibuat kurva pertumbuhan dengan spektrofotometer λ 600 nm dan dilakukan uji profil protein dengan metode SDS-PAGE. Hasil penelitian menunjukkan bahwa ketiga isolat uji tahan terhadap senyawa emas HAuCl4 sampai dengan konsentrasi 10 ppm dan semua isolat mempunyai protein yang berperan untuk resisten HAuCl4 pada berat molekul 38,9 kDa.

Gold or Aurum (Au) is one of the waste produced by the electroplating industry that can be toxic to some organisms, but some bacteria are able to tolerate. HAuCl4 is a chemical compound that contains Au. The research aims to determine the resistance of Gram positive bacteria to HAuCl4 and the difference of protein migration with electrophoresis using SDS-PAGE. The research begins with isolating resistance test of Bacillus Cereus S1, Lactobacillus sp. and Staphylococcus aureus using MSM medium broth + 2% glucose containing of HAuCl4 0,1 ppm; 1 ppm; 5 ppm; and 10 ppm. Cultures incubated for 24 hours. Then made a growth curve with the spectrophotometer λ 600 nm and tested protein profile with SDS-PAGE method. The results showed that the three isolates were resistant to gold compounds HAuCl4 up to a concentration of 10 ppm and all isolates had a contributing protein to HAuCl4 resistance at a molecular weight of 38.9 kDa.
