Abstract
Kanker payudara adalah kanker yang paling banyak diderita oleh wanita di dunia namun, pengobatan kemoterapi yang ada saat ini memiliki efek samping membunuh sel-sel normal pada pasien. Salah satu senyawa yang potensial sebagai senyawa antikanker baru adalah Trisindolina. Penelitian ini dilakukan untuk menguji sitotoksisitas senyawa Trisindolina 1 atau 5’-nitro [3,3’:3’,3”-terindolin]-2’one terhadap sel kanker payudara T47D dan untuk mengetahui aktivitas penghambatannya terhadap siklus sel T47D. Metode yang digunakan adalah microtetrazolium (MTT) assay untuk mendapatkan nilai Inhibitory Concentration 50 (IC50). Analisis siklus sel T47D dilakukan dengan metode flow cytometry menggunakan konsentrasi ½ IC50, IC50, dan 2 IC50. Doxorubicin digunakan sebagai kontrol positif. Nilai IC50 Trisindolina 1 terhadap sel T47D adalah 6,145 µg/ml, lebih tinggi dibanding nilai IC50 Doxorubicin yakni 0,145 µg/ml. Hasil uji siklus sel setelah penambahan Trisindolina 1 menunjukkan terjadinya cell cycle arrest pada fase G0-G1 serta meningkatkan akumulasi sel sub-G1 yang menunjukkan terjadinya peningkatan sel apoptotik. Mekanisme sitotoksik Trisindolina 1 terjadi karena adanya oksidasi indol menjadi indirubin yang menghambat aktivitas Cyclin Dependent Kinase (CDK).

Breast cancer is the most common cancer suffered by women in the world, but current chemotherapy treatment has normal cell killing side effect in patients. One of the compounds used as a new anticancer is Trisindoline. This study is conducted to test the cytotoxicity of Trisindoline 1 or 5'-nitro[3,3': 3', 3"-terindoline]-2'one against T47D breast cancer cells & to determine its inhibitory activity against the cell cycle of T47D cell line. The method used is Microtetrazolium (MTT) assay to obtain inhibitory concentration 50 (IC50) values. Doxorubicin was used as positive control. The cell cycle analysis of the T47D will be obtained by flow cytometry method using concentrations of ½ IC50, the IC50, & 2 IC50. The IC50 value of Trisindoline 1 against T47D cell line is 6,145 µg/ml, higher than Doxorubicin’s IC50 value which is 0,145 µg/ml. Cell cycle-assay after the addition of Trisindoline 1 indicates cell cycle arrest during G0-G1-phase & also increases cell accumulation in sub-G1 which shows the apoptotic cells. The cytotoxic mechanisms of Trisindoline 1 is due to the oxidation process of indole into indirubin that can inhibit Cyclin Dependent Kinases (CDKs) activities.
