Abstract

Diabetes melitus merupakan penyakit yang disebabkan oleh gangguan metabolisme yang ditandai dengan tingginya kadar glukosa dalam darah. Jumlah penderita penyakit diabetes melitus setiap tahunnya meningkat, tetapi pengobatan terhadap diabetes masih kurang maksimal. Hal ini karena obat yang dipakai masih menimbulkan efek samping. Oleh karena itu diperlukan senyawa obat lain yang minim efek samping. Studi literatur yang dilakukan mendapatkan bahwa trisindolina dapat dikembangkan sebagai lead compound senyawa antidiabetes. Penelitian yang dilakukan berhasil mensintesis dua trisindolina baru berupa 5',7'-dibromo-1,1',1''-tris(4-metilbenzil)[3,3':3',3''-terindolin]-2'-on (3a) dan 5',7'-dibromo-1,1''-bis(4-metilbenzil)-[3,3':3',3''-terindolin]-2'-on (3b). Trisindolina (3a) diperoleh dari reaksi 1-(4-metilbenzil)indola (5) dengan 5,7-dibromo-1-(4-metilbenzil)indoline-2,3-dion (4b), sedangkan trisindolina (3b) diperoleh dari reaksi 1-(4-metilbenzil)indola (5) dengan 5,7-dibromo indolin-2,3-dion (4a). Senyawa 1-(4-metilbenzil) indola (5) dan 5,7-dibromo-1-(4-metilbenzil)indoline-2,3-dion (4b) masing-masing disintesis terlebih dahulu dari reaksi N-alkilasi indola (6) dan 5,7-dibromoisatin (4b).

Diabetes mellitus is a disease that is caused by metabolic disorder and increasing level of glucose in blood. The number of patients with diabetes mellitus increases every year, but the treatment of diabetes mellitus is still not optimal. The drugs used still cause side effects. Therefore, new drug compound with minimal side effects is required. The literary studies found that trisindoline can be developed as a lead compound for antidiabetic compound. Two new trisindolines, in the form of 5',7'-dibromo-1,1',1''-tris(4-methylbenzyl)[3,3':3',3''-terindolin]-2'-one (3a) dan 5',7'-dibromo-1,1''-bis(4-methylbenzyl)-[3,3':3',3''-terindolin]-2'-one (3b), have been successfully synthesized in the study. Trisindoline (3a) was obtained from the reaction of 1-(4-methylbenzyl)indole (5) with 5,7-dibromo-1-(4-methylbenzyl) indoline-2,3-dione (4b), while trisindoline (3b) was obtained from the reaction of 1-(4-methylbenzyl)indole (5) with 5,7-dibromoindoline-2,3-dione (4a). 1-(4-Methylbenzyl)indole (5) and 5,7-dibromo-1-(4-methylbenzyl) indoline-2,3-dione (4b) are first synthesized from the reaction of N-alkylation indole (6) and 5,7 dibromoisatin (4b).
