Abstract
AntiMicrobial Resistance (AMR) menjadi masalah serious yang harus diselesaikan. Salah satu caranya dengan mengisolasi mikroorganisme yang mampu menghasilkan antibiotik. Sebanyak 70% mikroorganisme penghasil antibiotik dihasilkan oleh Actinomycetes. Actinomycetes mampu hidup pada lingkungan esktrem seperti Lumpur Lapindo. Isolasi bakteri Actinomycetes yang resistan terhadap ampicillin 0,125 ppm dari sampel Lumpur Lapindo pada koordinat Lintang Selatan: 70 31’46,23” dan Bujur Timur: 1120 42’36,83” telah dilakukan pada penelitin ini. Hasil seleksi menunjukkan bahwa isolat Actinomycetes berhasil diisolasi dan dimurnikan hingga koloni tunggal. Koloni tunggal isolat Actinomycetes A1 dilakukan pengujian pewarnaan gram, pengamatan kurva pertumbuhan, uji interaksi isolate bakteri terhadap jamur Aspergillus sp. dan Aspergillus fumigatus. Berdasarkan hasil pengujian pewarnaan gram diketahui bahwa isolat Actinomycetes A1 adalah bakteri gram positif dengan bentuk streptobasil. Berdasarkan kurva pertumbuhan dapat diketahui bahwa fase hidup isolat bakteri actinomycetes A1 adalah 34 jam. Uji interaksi isolat terhadap jamur Aspergillus sp. dan Aspergillus fumigatus menunjukkan hasil negatif.

AntiMicrobial Resistance (AMR) becomes a serious problem that should be solved. One of the methods is to isolate microorganism which capable of producing antibiotics. 70% antibiotic-producing microorganism is from Actinomycetes. Actinomycetes is able to live in extreme condition such as Lapindo Mud. In this study, the isolation of ampicillin-resistant actinomycetes 0.125 ppm from the Lapindo Mud sample in the coordinates of South Latitude: 70 31’46,23” and East Longitude: 1120 42’36,83” has been done. The results of the selection show that Actinomycetes isolates were successfully isolated and purified into single colony. Single colony isolate A1 was tested for gram staining, observation of growth curves, interaction isolate bacteria test against fungi Aspergillus sp. and Aspergillus fumigatus. In the gram staining, it is known that isolate A1 is a gram positive bacterium with a streptobacil form. Based on the growth curve, it can be seen that the life phase of isolates A1 is 34 hours. Test of isolate interaction of the fungus Aspergillus sp. and Aspergillus fumigatus shows negatife results.
