Abstract

Logam berat Pb, Cr dan Cd merupakan bahan beracun dan berbahaya. Proses pemulihan kontaminasi logam berat di lingkungan dapat menggunakan Bacillus cereus S1 yang terimobilisasi pada matriks alginat 2%. Isolat disubkultur pada Nutrien Agar miring dan diaklimatisasi pada Mineral Salt Medium (MSM). Uji resistensi dilakukan pada medium MSM Broth yang terpapar logam berat Pb, Cr dan Cd dengan konsentrasi 5 ppm. Akumulasi logam berat diukur dengan metode Inductively Coupled Plasma-Optical Emission Spectrometry (ICP-OES). Penelitian menggunakan Rancangan Acak Lengkap, perbedaan perlakuan diuji dengan ANOVA (α=5%) dan beda nyata diuji dengan Uji Tukey. Bacillus cereus S1 yang diimobilisasi pada matriks alginat 2% mampu mengakumulasi logam berat Pb, Cr dan Cd. Efisiensi akumulasi Pb hampir 100%, Cr 95% pada Batch I dan Batch II, Cd 49% pada Batch I dan 77% pada Batch II.

Heavy metals Pb, Cr and Cd are toxic and dangerous materials. The process of recovering heavy metals contamination in the environment can use Bacillus cereus S1 which is immobilized in the 2% alginate matrix. Isolate was subcultured in Nutrient Agar slant and acclimatized to Mineral Salt Medium (MSM). Resistance test used MSM Broth medium which was exposed to heavy metals Pb, Cr and Cd with a concentration of 5 ppm. Accumulation of heavy metals was measured by the Inductively Coupled Plasma-Optical Emission Spectrometry (ICP-OES) method. The research used Completely Randomized Design, the difference in treatment was tested by ANOVA (α = 5%) and the real difference was tested by Tukey Test. Bacillus cereus S1 which immobilized in the 2% alginate matrix was able to accumulate heavy metals Pb, Cr and Cd. Accumulation efficiency of Pb was almost 100%, Cr 95% in Batch I and Batch II, Cd 49% in Batch I and 77% in Batch II.
