Abstract

Konsorsium mikroba merupakan campuran populasi mikroba dalam bentuk komunitas yang mempunyai hubungan kooperatif, komensal, dan mutualistik. Secara alamiah, bakteri mampu berkomunikasi satu dengan yang lainnya. Antar bakteri dapat mempunyai hubungan sinergis jika substrat cukup dan tidak saling menghambat pertumbuhan. Pada penelitian ini telah dilakukan isolasi mikroba konsorsium yang resistan terhadap antibiotik ampisilin dengan konsentrasi 25 ppm dari lumpur Lapindo pada titik lokasi B1 (Lintang Selatan: 70 31’49,9” dan Bujur Timur: 1120 42’39,4”). Isolasi dilakukan dengan memilih isolat yang memiliki zona bening disekitarnya apabila berdekatan dengan koloni bakteri maupun jamur lain yang tumbuh dalam satu petri dish. Karakterisasi dilakukan dengan uji pewarnaan Gram, dimana hasil pewarnaan Gram menunjukkan bahwa komposisi konsorsium mikroorganisme A3 terdiri dari bakteri Gram-positif dan bakteri Gram-negatif berbentuk basil. Resistensi terhadap antibiotik dilakukan menggunakan metode dilusi broth untuk mengetahui minimum konsentrasi terhambatnya konsorsium A3 terhadap penambahan ampisilin. Pada uji dual kultur tidak didapati adanya interaksi penghambatan pada jamur Aspergillus fumigatus.

Microbial consortium is a mixture of microbial populations in the form of communities that have cooperative, commensal, and mutualistic relationships. Naturally, bacteria are able to communicate with each other. Between bacteria can have a synergistic relationship if the substrate is sufficient and between bacteria does not inhibit mutual growth. In this study, microbial consortiums that were resistant to ampicillin antibiotic with a concentration of 25 ppm from Lapindo mud at location points B1 (South Latitude: 70 31'49.9" and East Longitude: 1120 42'39.4") was isolated. Isolation was done by selecting isolates that have a clear zone around when close to other colonies of bacteria and fungi that grow in one petri dish. Characterization was carried out by Gram staining, where the results of Gram staining showed that the composition of the consortium of A3 microorganisms consisted of Gram-positive bacteria and Gram-negative bacilli bacteria. Antibiotic resistance was carried out using the dilution broth method to determine the minimum concentration of inhibition of the A3 consortium against the addition of ampicillin. In the dual culture test there was no inhibition interaction in the fungi Aspergillus fumigatus.
