Abstract
Penelitian ini bertujuan untuk mengetahui pengaruh konsentrasi substrat plastik dan lama waktu inkubasi terhadap kemampuan jamur dalam memecah rantai karbon plastik HDPE. Jamur yang digunakan dalam penelitian ini adalah Trametes polyzona (LM 1020), Aspergillus tereus (LM 1021), dan Aspergillus tamarii (LM 3010). Parameter yang digunakan dalam penelitian ini adalah pengukuran berat kering biomassa jamur, persentase degradasi (ED) plastik, analisis kadar protein crude enzim dengan metode Bradford, dan analisis aktivitas enzim alkana hidroksilase. Hasil penelitian menunjukkan bahwa persentase degradasi tertinggi (0,202±0,050%), kadar protein tertinggi (0,345±0,007 mg/ml), dan aktivitas enzim tertinggi (9,91±0,009 U/ml) terjadi pada isolat LM 1021 dengan konsentrasi substrat 8% dalam waktu inkubasi 50 hari. Untuk mempengaruhi waktu inkubasi terhadap degradasi plastik maka dilakukan analisis berat kering biomassa jamu. Berat kering biomassa jamur tertinggi 0,774±0,415 gram dengan konsentrasi substrat 6% dan dalam waktu inkubasihari ke-50 oleh jamur LM 3010.

This study aims to determine the effect of plastic substrate concentration and incubation time on the ability of fungi to break down HDPE plastic carbon chains. The fungi used in this study were Trametes polyzona (LM 1020), Aspergillus tereus (LM 1021), and Aspergillus tamarii (LM 3010). The parameters used in this study were measurements of fungal biomass dry weight, plastic degradation (ED) percentage, analysis of crude protein enzyme levels with Bradford method, and analysis of alkane hydroxylase enzyme activity. The results showed that the highest percentage of degradation (0,202 ± 0,050%), the highest protein content (0,345 ± 0,007 mg / ml), and highest enzyme activity (9,91 ± 0,009 U / ml) occurred in LM 1021 isolates with 8% substrate concentration in incubation time 50 days. To influence the incubation time of plastic degradation, an analysis of biomass dry weight of herbs was carried out. The highest dry weight of fungal biomass was 0,774 ± 0,415 grams with 6% substrate concentration and 50th incubation time by LM 3010 fungi.
