Abstract
Karsinoma hepatoseluler (HCC) merupakan kanker hati yang paling sering dijumpai. Agresivitas dan penyebarannya yang luas menyebabkan prognosis pasien yang buruk. Metode pengobatan seperti kemoterapi, radiasi, dan biopsi belum memberikan hasil maksimal bahkan memberikan efek samping pada sel normal, sehingga diperlukan metode pengobatan lain menggunakan bahan alam yang memiliki aktivitas antikanker yang relatif aman jika digunakan dalam pengobatan kanker. Salah satu bahan alam tersebut adalah senyawa Trisindolina 1. Penelitian ini menggunakan senyawa Trisindolina 1 dalam tujuh seri konsentrasi (1,563; 3,125; 6,25; 12,5; 25; 50; 100 µg/ml) yang diujikan pada kultur sel HepG2 dengan metode MTT-Assay (Microculture Tetrazolium Technique) dan analisis siklus sel menggunakan metode flowcytometry. Optical density diukur dengan menggunakan microplate reader, kemudian hasilnya dianalisa menggunakan regresi polnomial dan ditampilkan dalam bentuk nilai IC50. Hasil IC50 Trisindolina 1 adalah 9,247 µg/ml dan Doxorubicin adalah 0,803 µg/ml, sedangkan hasil analisis siklus sel dengan perlakuan Trisindolina 1 adalah HepG2 cell line arrest pada fase G0-G1 sedangkan Doxorubicin pada fase M1

Hepatocellular carcinoma (HCC) is the most common liver cancer. Aggressiveness and widespread spread cause a poor patient prognosis. Methods of treatment such as chemotherapy, radiation, and biopsy have not provided tangible results and even have normal side effects, determining the method of using natural ingredients that have anticancer activities which are relatively safe in the treatment of cancer. One of the natural ingredients is Trisindolina 1.This study uses Trisindolina 1 compound in seven series of concentrations (1,563; 3.125; 6.25; 12.5; 25; 50; 100 µg / ml) were tested on HepG2 cell culture using the MTT-Assay (Microculture Tetrazolium Technic) method and cell cycle analysis using the flowcytometry method. Optical density is measured using a microplate reader, then the results are analyzed using polynomial regression and displayed in the form of IC50 values.The result of IC50 Trisindolina 1 is 9.247 µg / ml and Doxorubicin is 0.803 µg / ml, while the results of cell cycle analysis with treatment of Trisindolina 1 HepG2 cell line arrest in the G0-G1 phase while Doxorubicin in M1 phase.
