Abstract

Kanker payudara adalah pertumbuhan sel payudara yang
tidak terkontrol karena abnormal gen yang mengatur pertumbuhan sel. Pengobatan kanker payudara seperti kemoterapi belum memberikan hasil maksimal, bahkan dapat memberikan efek samping pada sel normal, sehingga diperlukan alternatif senyawa lain yang memiliki aktivitas antikanker salah satunya adalah Trisindolina 1. Penelitian ini menggunakan senyawa Trisindolina 1 yang diujikan pada kultur sel 4T1 dengan metode MTT-Assay (Microculture TetrazoliumTechnique). Analisis siklus sel menggunakan metode flowcytometry. Optical density diukur dengan menggunakan microplate reader, dan hasilnya dianalisis menggunakan regresi polynomial dan ditampilkan dalam bentuk nilai IC50. Hasil penelitian menunjukkan bahwa Trisindolina 1 memiliki nilai IC50 sebesar 14,959 µg/mL dan Doxorubicin memiliki nilai IC50 sebesar 0,017 µg/mL. Pada uji flowcytometry Trisindolina 1 pada konsentrasi 14,959 µg/mL menunjukkan adanya persentase penghambatan akumulasi siklus sel pada fase G0-G1 dan Doxorubicin pada konsentrasi 0,017 µg/mL pada fase Sub G1. Berdasarkan hasil di atas, dapat disimpulkan bahwa Trisindolina 1 memiliki efek antikanker dengan menghambat proliferasi dan siklus sel 4T1.

Breast cancer is uncontrolled growth of breast cells because abnormal genes are subject to cell growth. Treatment of breast cancer such as chemotherapy has not provided maximum results, it can even give side effects to normal cells, so another treatment alternative that has anticancer activities is needed. One of them is Trisindolina 1. This study uses Trisindolina 1 compounds tested in 4T1 cell culture using the MTT-Assay method (Microculture TetrazoliumTechnique). Cell cycle analysis using the method of flowcytometry. Optical density was obtained using a microplate reader, and the results were analyzed using polynomial regression and obtained in the form of IC50 values. The results showed that Trisindolina 1 had an IC50 value of 14,959 μg / mL and Doxorubicin had an IC50 value of 0.017 μg / mL. In the Trisindolina 1 flowcytometry test at a concentration of 14,959 μg / mL the percentage difference in the percentage of cell cycle accumulation inhibition in the G0-G1 phase and Doxorubicin phases at a concentration of 0.017 μg / mL in the Sub G1 phase. Based on the above results, it can be concluded that Trisindolina 1 has an anticancer effect by inhibiting proliferation and the 4T1 cell cycle.
