Abstract

Astaxanthin merupakan salah satu antioksidan alami yang digunakan di bidang kesehatan dan akuakultur. Pembentukan antioksidan ini dipengaruhi oleh aktivitas enzim beta-karoten-ketolase yang disandi oleh gen BKT. Informasi gen BKT masih terbatas, sehingga eksplorasi mengenai gen BKT pada mikroalga beserta keanekaragamannya masih perlu untuk dilakukan. Tujuan penelitian ini adalah untuk menemukan isolat mikroalga yang berpotensi menghasilkan astaxanthin dan mengetahui hasil amplifikasi gen BKT pada mikroalga tersebut. Metode Sampling mikroalga pada penelitian ini adalah metode horizontal towing. Isolasi dilakukan di perairan hutan mangrove Kecamatan Sepulu, Kabupaten Bangkalan, Jawa Timur. Gen BKT pada isolat mikroalga diamplifikasi dengan PCR. Hasil dari penelitian ini adalah isolat murni mikroalga Chlorelloid yang didapat berpotensi memiliki gen BKT, hal ini didasari pada munculnya pita DNA pada gel elektroforesis hasil amplifikasi sekuens gen tersebut.

Astaxanthin is one of the natural antioxidants used in the health and aquaculture fields. Synthesis of astaxanthin is influenced by the activity of the Beta-carotene-ketolase enzyme encoded by the BKT gene. BKT gene information is still limited, so exploration of the BKT gene in microalgae and its diversity still needs to be done. This study aims to find isolates of microalgae that potentionally produce astaxanthin and to know the amplification result of BKT gene in isolated microalgae. The sampling method for microalgae in this study is the horizontal towing method. Isolation was carried out in mangrove forest waters of Sepulu Sub-district, Bangkalan. The gene of microalgae obtained were amplified using PCR. The result of this research is isolates of Chlorelloid microalgae potentionally has BKT gene, this result based on the emergence of DNA band on electrophoresis gel after the sequence of BKT gene being amplified.
