Abstract
Pada penelitian ini telah dilakukan penumbuhan jamur Penicillium sp. B96 menggunakan metode fermentasi media cair selama 7 hari dengan variasi sumber karbon yaitu glukosa, fruktosa, dan galaktosa. Hasil fermentasi disentrifus untuk memisahkan supernatan dan endapan (pelet). Supernatan yang di dalamnya terdapat kandungan zat metabolit sekunder digunakan untuk uji antibakteri dengan metode difusi agar pada kertas dis terhadap dua bakteri uji yaitu Staphylococcus aureus dan Escherichia coli. Berdasarkan kurva pertumbuhan yang diperoleh, variasisumber karbon glukosa dan fruktosa mencapai fase stationer yang dimulai pada hari keempat, sedangkan galaktosa dimulai pada hari keenam. Uji aktivitas antibakteri pada bakteri S. aureus (Gram-positif) dan E. coli (Gram-negatif) masing-masing menunjukkan hasil diameter zona hambat optimum pada variasi glukosa hari ketujuh sebesar 11 mm dan 10 mm.

In this study, Penicillium sp. B96 was grown using a liquid media fermentation method for 7 days with a variety of carbon sources, namely glucose, fructose, and galactose. The fermented product is centrifuged to separate the supernatant and sediment (pellets). Supernatants which contain secondary metabolites are used for antibacterial testing and using agar diffusion method on paper dis for two test bacteria, namely Staphylococcus aureus and Escherichia coli. Based on the growth curve obtained, variations in the sources of carbon glucose and fructose reached the stationary phase which started on the fourth day, while galactose starts on the sixth day. Antibacterial activity tests on S. aureus (Gram-positive) and E. coli (Gram-negative) bacteria each showed the optimum inhibition zone diameter on the seventh day glucose variation of 11 mm and 10 mm.
