Abstract

Pertumbuhan tanaman kentang (Solanum tuberosum L.) secara in vitro dipengaruhi oleh ZPT dan Pencahayaan. Penelitian ini bertujuan untuk mengetahui pengaruh penyinaran dan kombinasi Zat Pengatur Tumbuhan terhadap pertumbuhan bibit kentang varietas Granola Kembang. Penelitian dilaksanakan di Laboratorium Kultur Jaringan Tanaman CV. Muria Sari Bumi, Pesanggrahan Batu pada bulan Desember 2018 – April 2019. Eksplan digunakan umbi kentang yang ditanam di media MS dengan perlakuan kombinasi NAA+BAP: NAA 0,01 mg/l + BAP 0,01 mg/l, NAA 0,02 mg/l + BAP 0,02 mg/l, NAA 0,03 mg/l + BAP 0,03 mg/l, NAA 0,04 mg/l + BAP 0,04 mg/l, NAA 0,05 mg/l + BAP 0,05 mg/l. Kultur diinkubasi pada suhu 20°C dengan pencahayaan menggunakan lampu LED putih dan merah biru dengan lama penyinaran 12 jam. Penelitian menggunakan Rancangan Acak Lengkap 2 Faktorial dan perlakuan diulang sebanyak 5 kali. Data yang dihasilkan dianalisa dengan ANOVA two way, menggunakan software SPSS. Hasil penelitian menunjukkan perlakuan NB4P1 (NAA 0,04 mg/l + BAP 0,04 mg/l dan Pencahayaan Putih) yang memiliki kenaikan tinggi planlet paling tinggi yaitu 18,36 cm. Parameter jumlah daun didapatkan hasil tertinggi pada perlakuan NB5P1 (NAA 0,05 mg/l + BAP 0,05 mg/l dan Pencahayaan Putih) yaitu 18 helai daun kentang dan pada parameter jumlah akar didapatkan data tertinggi pada perlakuan NB5P2 (NAA 0,05 mg/l + BAP 0,05 mg/l dan Pencahayaan Merah) yaitu 14 helai akar kentang.

In vitro growth of potato plants (Solanum tuberosum L.) is influenced by plant growth regulators (PGR) and lighting. This study aims to determine the effect of illumination and combinations of Plant Growth Regulators on the growth of Granola Kembang variety potato seedings. The study was conducted at the Plant Tissue Culture Laboratory of CV. Muria Sari Bumi, Pesanggrahan Batu from December 2018 to April 2019. Potato tubers were used as explants grown on MS medium with combination treatments of NAA+BAP: NAA 0.01 mg/l + BAP 0.01 mg/l, NAA 0.02 mg/l + BAP 0.02 mg/l, NAA 0.03 mg/l + BAP 0.03 mg/l, NAA 0.04 mg/l + BAP 0.04 mg/l, NAA 0.05 mg/l + BAP 0.05 mg/l. Cultures were incubated at 20°C with illumination using white and red-blue LED lights with a photoperiod of 12 hours. The study used a 2-Factorial Completely Randomized Design, and treatments were repeated 5 times. The generated data were analyzed using two-way ANOVA with SPSS software. The results showed that the NB4P1 treatment (NAA 0.04 mg/l + BAP 0.04 mg/l and White Lighting) had the highest increase in plantlet height, which was 18.36 cm. The parameter of the number of leaves obtained the highest result in the NB5P1 treatment (NAA 0.05 mg/l + BAP 0.05 mg/l and White Lighting) with 18 potato leaves, and for the parameter of the number of roots, the highest data was obtained in the NB5P2 treatment (NAA 0.05 mg/l + BAP 0.05 mg/l and Red Lighting) with 14 potato roots.
