Abstract
Kulit ikan patin (Pangasius hypophthalmus) merupakan produk samping industri fillet ikan yang berpotensi menjadi alternatif bahan baku pembuatan gelatin halal. Bahan baku dan faktor fisik-kimia dalam proses ekstraksi berpengaruh terhadap rendemen gelatin yang dihasilkan. Penelitian bertujuan untuk optimasi ektraksi kulit ikan patin (kip) pada ukuran yang berbeda. Metode yang diuji meliputi: Analisis kolagen mengguankan histologi kip dengan pewanaan Mason Trichome; Analisis optimasi pretreatment-ekstraksi menggunakan Placketburman dan penetuan nilai optimum varibel ekstraksi menggunakan Response Surface Methodology (RSM), uji rheological and uji biuret untuk mengvaluasi kualitas gelatin. Ikan patin yang digunakan dalam penelitian terdiri dari 2 ukuran yang mewakili ukuran kecil (38,9 ± 0,5 cm) dan ukuran besar (48,6 ± 1,94 cm). Hasil penelitian menunjukkan bahwa kulit dari ikan berukuran kecil menunjukkan densitas kolagen (13180,45 rawintdensity); rendemen gelatin (50.83%); kekuatan gel (223 - 466 bloom) dan viskositas (6.53-7.4 cp) yang lebih tinggi daripada ikan besar (densitas kolagen=12835.21 rawintdensity; rendemen gelatin =32.35%; kekuatan gel= 154-199 bloom, viskositas=3.60–3.66 cp. Kadar protein total pada gelatin dari kulit ikan patin yang berukuran kecil maupun besar >85% . Berdasarkan penelitian disimpulkan bahwa kulit ikan patin memiliki potensi tinggi sebagai bahan gelatin halal, terutama dari ikan berukuran kecil.

Skin of Pangasius hypophthalmus is a side product of fish fillet industry which has the potential to be an alternative raw material for making halal gelatin. Raw materials and physical-chemical factors in the extraction process affect the yield of the gelatin. The study was aimed to optimize the extraction of skin P. hypophthalmus at different sizes. There were several analysis included: histological analysis of collagen using Mason's Trichome staining; Optimization of pretreatment-extraction analysis using Placket-burman and determination of the optimum value of extraction variables using Response Surface Methodology (RSM); rheological test and biuret test to evaluate the quality of gelatin . The catfish used in the study consisted of 2 sizes representing a small size (38.9 ± 0.5 cm) and a large size (48.6 ± 1.94 cm). The results showed that the skin of small fish showed collagen density (13180.45 rawintdensity); gelatin yield (50.83%); gel strength (223 - 466 bloom) and more viscosity (6.53-7.4 cp) than large fish (collagen density = 12835.21 rawintdensity; gelatin yield = 32.35%; gel strength = 154-199 bloom, viscosity = 3.60–3.66 cp The total protein content of the gelatin from the skin of P. hypophthalmus was in value of > 85%. The research found that the skin of P. hypophthalmus has high potential as a halal gelatin material, especially from the small-sized fish.
